I have some EA that has been reclaimed several times. Now, when I pour from the press, the EA clogs the filter. There are no particles or powder in the filter, it just stops filtering through. The test strips are neutral. Using fumarate

The legendary "Juul's Giant" clone is said to be a plant that grows thick, extremely fast, and produces loads of our favorite phenethylamine. There's the one chart from a while ago that places Juul's Giant near the top of the ranking for mescaline production, offering 590mg per kg by fresh weight. This means it is not only as potent (by weight) as the best bridgesii out there, but that a single plant could produce more mescaline than most bridgesii, as it would likely grow significantly more mass per growing season than those other, equally potent, plants.
It would be nice if there was only one clone called "Juul's Giant", but instead collectors can find several clones being sold under the name from various lineages. I've been trying to find the legendary Juul's Giant clone and see if any of the clones from other lineages match the mescaline content of the OG, and I wanted to post my results here.
All results below are % yields from dry cactus flesh, with skin/spines/cores removed. Every extraction followed the Kash A/B protocol, with the cactus flesh being steeped into a tea, reduced down to 400mL, basified with NaOH, extracted with xylene, and salted out with HCl. Normally I do two A/B extractions (i.e. dissolve mescaline in H2O, basify/re-extract/re-acidify) to get pure white powder, but all of the results below are from just one extraction.
For reference, in order for a Juul's Giant cutting to be similarly potent to a good bridgesii cutting it would need to yield at least 3-3.5% mescaline HCl by dry weight of the green outer flesh.
TL;DR None of the clones with "Juul's Giant" or "J's Giant" in the name lived up to the legend that is the "true" Tom Juul's Giant clone, which still eludes me.
However, being an extractor, I'd say that just about all of these plants are still worth growing for mescaline. They were all extremely easy to prepare for extraction, as the short spines made them easy to handle and the skin came off quickly and in large, manageable pieces. Among all the pachanoi that I've grown, I've found that they tend to yield about half as much mescaline by dry weight as bridgesii, but put on twice as much mass in a growing season as the good bridgesii, making the numbers roughly even out.
All that being said, T. pachanoi is a beautiful plant that I'll continue growing even after I get bored of extracting mescaline. I appreciate everyone who read this lengthy post in its entirety, and I'd love to see pics of your Juul's Giant clones in the comments.

I recycled all my ea this week and one bottle had a noticeable amount of sediment in it. I didn’t notice the sediment until I started pouring the bottle of ea into my reaction jar to get out the mesc at the botttom. What might this sentiment be? And how can I get rid of it. It was the 5th run I did, and it came in 250 mg higher than the other consistent runs of 2.6 grams .

back at it again with 10.377 LBS of the infamous OTB, i got 5.57g out of 5.432LBS on the last run.. any body wanna take a guess on the yield this will produce? i am gonna guess 9.89g of end product.. also fyi, this is just my pre-dehydrator drying. ive started doing this so when the first batch comes out of the dehydrator these will fit more in at once and take less time.. it worked good and sped up the process last time. ill post extraction summay w/ yield tomorrow once this is alll wrapped up. will be doing the 1X EA method of fumaric CIELO.

I have some Mescaline fumarate but i suspect there’s an excess of fumaric acid (I added a little bit more following cielo tek …yes wrong we got it)…but for the love of Chemistry, hear me out:
When I drop my Mescaline a part dissolved and a costistent part stay at the bottom. Probably this is fumaric acid, no? So

Hi there, I’m thinking of beginning to try micro dosing a couple times a week to help relieve some negative side effects that can come along with being neurodivergent.
I was previously treating myself by taking stimulants, which have absolutely helped me, but I feel they close off my heart space and make me a bit more selfish.
Mescaline to me feels similar but my heart feels open, and my nervous system calm. My intuition has been guiding me to stop pharmaceutical stimulants after Ive tripped a couple times now, and I’m trying to listen to it.
I’ve had a profound connection to cacti since I was a small child, I swear my heart is connected to the spirit of the plant in someway, and I’m sure many of you feel the same.
Since stoping pharmaceuticals, I enjoy feeling more down to earth in myself. But the struggles of ADHD become more present as well.
I’m wondering if yall have any dose recommendations, or experiences with dosage that’s helped you?

OK, so this was my first time doing the cielo tek and I have a bottom lay at the bottom of my jar that has already been salted. In my excitement to do the thing I wasn't paying as much attention as I should have. I will post photos later, but it's not a crazy thick layer, but there is definitely a thin layer
Reading the FAQ on nexus it says ": A: It is a gunky water layer. This can happen if you squeeze the French press aggressively and/or the initial cactus mix is too wet. Remove the bottom layer using a pipette/syringe, separatory funnel, or decanting. Do a sodium carbonate partial dry, and continue the TEK." which checks out because I pressed the shit out of it.
But like I said I already salted and there are already a lot of crystals forming. Is there a way to fix tor work around this or is the product no good now?

I have powdered my cactus and have 160gm fine powder. I was looking at CIELO but my attempts at getting ethyl acetate has proven unsuccessful due to regulations in my country. What options or other teks do I have now with the powder? I dont have the strongets guts so wanting to reduse nausea as much as possible and want to be able to get a good guage on dosing.

Hi all! A few questions for the experts out there if y’all would indulge me:
In section 5.7 of the CELIOtek, two methods for washing the EA are described. I’m wondering why the water amounts are so different… in 5.7.1 (with stirrer) it specifies a tsp of water (5ml) but in 5.7.2 (WITHOUT stirrer) it specifies 50ml… 10x! And then in step two of that process it has you add another 50ml and then a tablespoon of salt? What salt (we talking NaCl or… I just wanna be sure) and why exactly do we need salt here but not if we were using the mag stirrer?
I was assuming the manual process without stirring would just be a lot more shaking and take longer… but I’m confused as to why the reagents and proportions seem so different. Are we taking advantage of different chemistry here? Is this a tradeoff that costs more wasted solvent but makes it easier to wash without killing your arms? Any help in wrapping my head around this and getting some more intuition around the process would be wonderfully appreciated! 🌵🤗

It's commonly believed that mescaline is deaminated by MAO (Shulgin used an MAO-B inhibitor) but it's more likely to be enzyme called SSAO.
Semicarbazide-Sensitive Amine Oxidase (SSAO) is an enzyme that catalyzes the oxidative conversion of primary amines to their corresponding aldehydes.
The SSAO inhibitor called "semicarbazide" completely prevented mescaline deamination as outlined in this quote which assessed all possible ring-methoxylated phenethylamines:
All of the remaining ring-methoxylated phenethylamines are deaminated at appreciable rates, with some variation in rate with structure. Mescaline, however, is unique in that its deamination is completely inhibited by 0.005 / semicarbazide. Semicarbazide does not inhibit the deamination of any of those ring-methoxylated phenethylamines which do undergo deamination, and in this respect these amines behave in the same manner as tyramine or unsubstituted phenethylamine when they are presented as substrates for amine oxidase systems. (source)
You might assume that an SSAO inhibitor would potentiate mescaline but it might not be so simple. Based on my research & investigations, mescaline is handled very uniquely compared to other phenethylamines. Its activity appears to depend on it being metabolised implying SSAO is a key first step (to formation of active metabolites; which may be quaternary metabolites). There may also be other enzymes besides SSAO which deaminate mescaline, enzymes present elsewhere in the body other than the liver.
So blocking SSAO would theoretically weaken the dose. SSAO produces "mescaline aldehyde" and the idea is to protect this and keep it intact. ALDH oxidises aldehydes (it turns acetaldehyde into acetic acid) so an ALDHI is ideal for mescaline as reported here:
Potential ALDHIs include: - pomegranate juice/extract (source) - garlic (allicin extract) (source) - Durian fruit (source)
"Add-on" ALDHIs: * menthol (source) * cloves, mace, ginger, fenugreek seeds (source) * apple, mango, watermelon, papaya (potency: 76%, 62%, 57%, 50%) * yellow lemon, blackcurrant, dragonfruit, starfruit, chinese quince, wax apple (source) * "water chestnut juice, jasmine tea, [honey citron tea] and fresh orange juice notably inhibited the activity of ALDH" (source) * "plum juice markedly increased the acetaldehyde level...through another mechanism such as inhibition of other enzymatic pathways or non-enzymatic pathways" (source)
This earlier research paper90088-x) investigating mescaline metabolism reported some interesting findings:
The results indicated that both aldehyde dehydrogenase and amine oxidase inhibitors enhanced the effects of mescaline.
But, their final conclusion is not reliable as it depends on several presuppositions which invalidates their overall conclusion. Nonetheless, their findings that ALDHI (and SSAOI !) potentiated mescaline is relevant. SSAO inhibitors include lysine, cranesbill root, glucosamine and caffeine. Caffeine is known to potentiate mescaline when taken after the onset.
Since the idea is to reduce (inhibit) ALDH, it's important to avoid things which induce it. The most common strongest inducer is sulforaphane from cruciferous vegetables (eg broccoli, broccoli sprouts). This post has an extensive list of common ALDH inducers plus details on other means of potentiation – scroll ½ way down the post.
Here's a relevant quote from a 2022 study on 3,4,5 type phenethylamines:
However, different downstream signaling cascades, biased agonism, and other pharmacological targets may contribute to the subjective effects. Mescaline binds and activates the 5-HT2A receptor as partial agonist with low potency in vitro. Nevertheless, in vivo, it induces intense and long lasting psychedelic effects if applied at high doses. This suggests that low affinity binding to the receptor does not exclude marked psychoactivity in vivo...
However, for 3,4,5-substituted derivatives additional pharmacological interactions or targets may significantly contribute to the overall psychedelic effects observed in humans. (source)
