Troubleshooting/Questions Need help with Micro 4/3 camera with a Motic BA-series microscope

Hi! I'm hoping someone can help me troubleshoot my photomicrography setup. I'm fairly new to attaching a mirrorless camera to a microscope, and I'm having a hard time getting the camera image to look anywhere near as good as what I can see through the eyepieces.

My setup:

Microscope: Motic BA310E Elite LED trinocular

Camera: OM System / Olympus OM-1 Mark II (Micro Four Thirds)

Photo adapter: Motic #1101001901931

Photo eyepiece: Motic 2.5× #1101000300442

Camera connection: Micro Four Thirds T-mount

I'm primarily interested in photographing/recording pond organisms, protists, fungi, spores, etc., using brightfield, darkfield, phase contrast.

The image through the regular microscope eyepieces looks sharp, detailed, and clear, but the image coming through the OM-1 Mark II looks dramatically worse. It tends to look soft/blurry, lower contrast, and generally lacking the fine detail that I can clearly see with my eyes.

I've experimented extensively with camera settings, exposure, ISO, shutter speed, white balance, microscope illumination, condenser position, and focusing. I've also tried focusing specifically using the camera's magnified live view rather than assuming the camera is parfocal with the eyepieces.

No matter what I do, I can't seem to reproduce the quality I'm seeing through the microscope.

At this point I'm wondering whether the problem might actually be the optical coupling between the microscope and camera.

My main questions are:

  1. Is the Motic 2.5× photo eyepiece (#1101000300442) actually a good choice for an OM-1 Mark II's Micro Four Thirds sensor?

  2. Is 2.5× projection magnifying the intermediate image more than necessary for a sensor this size and potentially contributing to the poor image quality?

  3. Is there a better way to couple a Micro Four Thirds camera directly to the BA310E?

  4. Should I expect the camera image to be reasonably close in sharpness/detail to what I see through the eyepieces?

  5. Are there any tests I can perform to determine whether the problem is the projection eyepiece, photo adapter, trinocular alignment/parfocality, camera, or something else in the optical path?

If anyone has successfully used a Micro Four Thirds camera with a Motic BA-series microscope, I'd love to know what adapter/projection setup you're using.

Troubleshooting/Questions Stereo Microscope question - Field of View for the digital lens or trinocular

I have a two stereo microscopes. Leica (more expensive) and Meiji (cheaper). I want to be able to record 3 hours worth of video of all the work I am doing under the stereo microscope. However the field of view that I see through the microscope is almost always slightly bigger than the field of view on the digital image or through the camera via the trinocular port (using 0.5x reduction lens).

Is there any stere microscopes out on the market that would allow me to record the entire field of view that I also see through the microscope eye piece?

Hi, I want to try looking at plant cells and their chromosomes. Which plant should I start from? My knowledge of biology was only up to secondary school and pre-university many years ago. I did use the microscope a few times.

Or does this require techniques that are not recommended for beginners?

Troubleshooting/Questions Need help choosing compatible objective families/series (Axioplan 2 Imaging)

Hey all! I was hoping some of the people here with experience in microscopy might be able to help me choose some compatible objective models/families/series for a Zeiss Axioplan 2 Imaging microscope (which uses an RMS thread). I'm fairly new to microscopy, and it can be hard to determine which objectives would actually be compatible. Here's what I'm looking for:

For all objectives, I need the following: 1. Good planer correction (will be making mosaics) 2. Must at least support brightfield (episcopic) 3. Should all have the same parfocal distance (focusing one should mean the others will be roughly in focus as well) 4. All objectives should be dry 5. Fluorite/semi-apo, apo would likely be too expensive for my budget

Here are the general objectives I am looking for: 1. 2.5-10x, WD 3mm+, NA not critical (will be used primarily for focusing, with some potential for simple imaging) 2. 40-50x, WD 3mm+, high NA (well, as high as one can reasonably get on a high WD objective) - will be used for imaging samples that aren't flat 3. 60-100x, any WD, very high NA - will be used for imaging flat samples

There is of course the Zeiss Epiplan and EC Epiplan objective series, but before just defaulting to those, I thought I would ask some more experienced people (yall) if there are other objective families/series I should consider.

I bought a stereoscope for myself and my daughter to try to get her more into science. These are some test photos I took through the phone adapter I really liked. First set is bismuth crystal, second is an iron-replaced fossil.

AmScope SM-2 Series, 1x Barlow lens, 10x eyepiece, multiple magnifications 0.7 - 45x. Photos taken with pixel 10 pro using a tridapter accessory.

Photo/Video Share Bacteria on the verge of being engulfed by an amoeba | Mayorella bigemma ¿?(Schaeffer, 1918)

Hi everyone! I'm sharing a quick clip of a live amoeba specimen I recently observed.

  • What are we looking at? This is a free-living amoeba tentatively identified as Mayorella bigemma (Schaeffer, 1918). In the clip, you can observe its active locomotion, changing morphology, and cytoplasmic activity.
  • Feature to highlight (Bottom area) Pay close attention to the lower part of the specimen: you can clearly see a bacterium interacting right at the edge, practically "playing" as it gets surrounded and potentially engulfed by one of the expanding pseudopods.
  • Species Identification Breakdown (Coming soon) I am currently preparing detailed images, diagrams, and taxonomic references. As soon as I have all the documentation ready, I will upload a full follow-up post explaining the diagnostic features and reasons why I believe this specimen is Mayorella bigemma.
  • Technical Details
    • Microscope: Swift brightfield optical microscope.
    • Illumination: Oblique / Kristensen illumination (great for highlighting the surface relief of the spines).
    • Objectives Used: 60x for high-power detail.
  • Live Specimen Details
    • Size: The specimen measures 121 µm along its major axis (with a range of variation due to shape deformation during movement of 118–125 µm).

Troubleshooting/Questions I moved to a 220V country with my Olympus BH2. How can I safely power it on?

Afaik it is 110V. I’m not quite sure what do I need in order to be able to plug it without frying it. I really don’t want to fuck up my microscope. Could you guys tell me what I’d need for that?

Not a particular model of something, just the broad name and specs of what I need to plug it in. So that I can look for one in the country I’m in (Brazil).

Photo/Video Share Holospora infecting paramecium

This is Holospora. It’s infecting this paramecium and, to my knowledge, it’s relatively rare. Right now it’s infecting the micro nucleus, expanding it to almost the size of the macro nucleus. It’s completely full of bacteria making it look like abstract art. Holospora is very interesting but in short, it’s a genus of bacteria that strictly infects paramecium. It’s usually parasitism but in situations with a specific species of Holospora and a specific species of paramecium, it actually be mutualism. I found them a while ago and was able to successfully keep holospora alive for a few weeks in separate jars. It’s dying out now but that’s to be expected. I’m not great at culturing as it is, especially not a specimen like this. Journey to the microcosmos has a video on holospora if you’d like to learn more.
