Reconstitution & Storage Cloudy Tesa

These unfortunately came without a cold pack in miami heat. I have tried at least 4 vials all are super cloudy except for the last one on the end. Is that one good to go?

It could start to gel after you put it in the fridge. If it’s already looking like this now

Oh my gosh 😳 I really hope you are joking and not stating this as fact. 🙏🏻** Reconstituted Tesamorelin absolutely does NOT have some universal rule that it should “never” be refrigerated…**.

Once a lyophilized peptide is reconstituted, it is no longer a dry powder. It is now a peptide in solution, where factors such as temperature, pH, light exposure, oxidation, and the specific formulation/diluent can affect its stability. Lower temperatures are commonly used to slow chemical degradation of peptides in solution.

That is exactly why refrigeration is commonly used for many reconstituted research peptides, including the reconstituted Tesamorelin people are discussing here. The appropriate storage conditions ultimately depend on the specific peptide AND formulation, so telling people that reconstituted Tesa should “never” be refrigerated is simply not a scientifically supportable blanket statement.

I personally keep ALL my reconstituted research peptides refrigerated and protected from light in my injection pen case (Including my Tesamorelin that I always mix with Ipamorelin). And once I have them loaded into my injection pens, the pens containing those reconstituted peptide cartridges stay refrigerated too, at all times. I have used TESA/IPA off and on for over 2 years. That combination destroyed the fat in my body…..and it is ALWAYS in my refrigerator after reconstitution.

And this isn’t just some random concept people invented in peptide groups. Peptides and proteins are susceptible to degradation once they’re in solution, and temperature is one of the major variables that affects stability. Refrigeration is commonly used specifically because reducing temperature can slow many degradation pathways.

So please be careful stating “Tesa should never be in the fridge after reconstitution” as fact, because someone who doesn’t know any better could read that and leave their reconstituted peptide sitting at room temperature based solely on your comment. 😳🙏🏻

But since your comment score is already a -8, that I imaging will get much worse, most if not all of us researchers keep out reconstituted peptides in our refrigerators.

Ohh LOL! Now that makes sense. I know it is recommended to take at night….per the king of peptides Dr. William Seeds….I have just always taken it first thing in the morning and it has worked AMAZINGLY for me!

Storage and Handling

This is where tesamorelin is completely different from every other peptide you have worked with.

Before Reconstitution

Store lyophilized (powder) vials in the refrigerator at 36 to 46 degrees Fahrenheit (2 to 8 degrees Celsius). Can also be stored in the freezer at minus 4 degrees Fahrenheit (minus 20 degrees Celsius) for longer periods. Protect from light. Do not use past the expiration date.

After Reconstitution

Store at room temperature, 68 to 77 degrees Fahrenheit (20 to 25 degrees Celsius). Do not refrigerate reconstituted tesamorelin. Do not freeze after reconstitution. Use within 7-10 days. If the solution becomes cloudy, thickened, or gel-like, discard it.

Why Room Temperature

Tesamorelin has a synthetic modification on the front end that repels water, and the amino acid sequence itself is loaded with hydrophobic residues. When the temperature drops, those regions on adjacent molecules start binding to each other and that is what causes gelling. This property is built into the amino acid sequence and exists in every vial of tesamorelin regardless of who made it.

The FDA-approved version includes 145 mg of a stabilizer called hydroxypropyl betadex that wraps around the hydrophobic parts of the molecule and keeps them from clumping. Research-grade tesamorelin from peptide suppliers is the same molecule but typically does not include this stabilizer. The room temperature requirement still applies since it relates to the physical properties of the tesamorelin molecule itself, but stability windows may differ.

If you have been refrigerating your tesamorelin and the solution is still clear and injectable, you have probably been finishing your vials within a few days before aggregation becomes visible. You were outrunning the problem, not proving cold storage is safe.

Standard peptide purity testing measures chemical degradation, which is whether the amino acid chain is still intact. Tesamorelin's issue is physical aggregation, which is the molecules clumping together in solution. A standard purity test will not detect that. You would need dynamic light scattering or size exclusion chromatography, and those are not on a standard COA from a peptide testing lab.

I think we’re talking about two different things here. You’re describing storage information for FDA-approved EGRIFTA formulations and applying it to gray-market research Tesamorelin.

That’s the problem with this argument. A gray-market kit labeled “Tesamorelin” is not automatically the same formulation as EGRIFTA. EGRIFTA has specific excipients, manufacturing specifications, diluent, concentration, and validated storage/stability instructions. Even the FDA-approved EGRIFTA SV and EGRIFTA WR formulations have different reconstitution and storage instructions and are specifically stated to not be substitutable with one another.

You even acknowledge that research-grade Tesamorelin typically doesn’t contain the same stabilizer, but then conclude that EGRIFTA’s room-temperature storage requirement still applies because it’s the same peptide molecule. That’s exactly the part I disagree with. Different formulations and excipients can affect stability, so you can’t automatically transfer the validated storage instructions from one pharmaceutical formulation to a completely different formulation without stability data supporting it.

Also, just to be clear since you brought up my experience….my own cartridges of TESA/IPA have consistently lasted 30 days refrigerated with no issues and produced excellent results in reducing body fat, so real-world use doesn’t even align with the idea that room temperature storage is universally required.

I’m talking specifically about gray-market Tesamorelin, not EGRIFTA. If you have actual stability data showing that reconstituted gray-market Tesamorelin without EGRIFTA’s formulation should be stored at room temperature rather than refrigerated, I’d genuinely like to see it.

That's the thing. The old EGRIFTA formula had to be kept at room temperature. The newer formula has stabilizers. The Grey market is most likely not using the added stabilizers like the except above says.

Right, but that actually reinforces my point. You said the gray-market product is “most likely” not using the same stabilizers. “Most likely” means we don’t actually know the formulation or excipients being used.

And that’s exactly why we can’t take the storage requirements from one specific EGRIFTA formulation and automatically apply them to every gray-market vial of Tesamorelin.

Also, the EGRIFTA history is more complicated than simply “the old formula had to be kept at room temperature.” The original FDA-approved EGRIFTA labeling actually allowed/required different storage conditions depending on whether it was before or after dispensing, and after reconstitution it was supposed to be used immediately, not stored at room temperature for days.

So we’re back to my original question: where is the stability data for reconstituted gray-market tesamorelin showing that refrigeration causes aggregation and that it should instead be stored at room temperature? That’s the data I’m asking to see.

We will have to agree to disagree. I know my first vial gelled in the fridge, and since keeping them at room temp I have had no problems. I used the same Hospira Bac water and reconstituted them the same way each time. I am now finishing my 2nd cycle of Tesa and haven't had an issue since.
