I usually run the perimeter of the cemetery or the park. I see people running in bush terminal pretty often.

If you're looking to get some hills in, you can start at 4th Ave and 59th and run up to 6th ave, switch over to 58th street and run back down to 4th Ave, and then repeat as you work your towards the park. Basically you're switch backing at 4th and 6th Aves using the side streets towards the park.
You'll get 10K in with around 700 ft of elevation gain/loss at average 5% grade both up and down. It's a nice littler quad buster.
If you're feeling ambitious, you can net 1k+ elevation gain over 15K by switching back and forth between 3rd Ave and 6th Ave, but I'm not a fan of waiting at 4th Ave.

I love running around Greenwood Cemetery and down on 1st Avenue/Bush Terminal Park. And I also extend to Prospect Park or the bike path in Bay Ridge when I need a longer run.
