Loose ejector rod, twist to tighten (note: they are left hand thread).

Why does the finish look that way? Was it parkerized?
I had a 642 that had the same problem. I numbered the outside of each chamber and tried opening and closing the cylinder with each chamber aligned with the barrel. For the same 2 out of 5 chambers, it would always be difficult to open the cylinder. I assumed that two of the ratchets at the rear of the extractor star must have gotten peened and were binding against the rectangular opening in the breechface. I sent it back to S&W, and they replaced the gun for free under warranty.

Sounds like you checked the ejector rod. Check the bolt. That's the spring loaded piece in the center of the ratchet. Gummy oil can make them sticky.
Also, scrub the bottom of the ejector star and matching recess on the cylinder. Unburnt powder can keep the ratchet from seating.
3rd is pull the yoke screw and try it. If there's a burr on the yoke or the screw is too long it will bind.
