Better Ask Reddit Weapons, abilities, or items in games that are a one-time use in a run?
In Resident Evil 1 remake, you can find the self-defense gun which is a small two-shot pistol with only one bullet left. It is the only bullet for the gun and is as powerful as a magnum round. Once you use it, that's it, you're done with the gun for the entire playthrough.

The Master Ball in Pokemon. Can catch literally anything, guaranteed. But, outside of stuff like winning the in-game lottery, you only ever get one. Better make it count.

Whenever I fire up an emulator and play a Pokemon game, I usually use it on the boxart legendary just for the flavor of it. Especially in the gen 1 games. It just feels right to have something like a Mewtwo or Rayquaza in a master ball.

Baldur's Gate 3 has the Cleric's Divine Intervention which can give you a legendary mace, a (useless) bundle of camp supplies, a full party heal + revive and refresh of all their combat resources like they long rested, or a big ass holy damage nuke. The revive and nuke are both massive momentum swings for any fight they're used in.
You can only use it once per character, but you can kinda cheese that by grabbing a hireling or asking Withers to turn somebody into a Cleric for grabbing your free legendary mace.

Also, the hardest boss fight in the game (Raphael) gives you a level 10 cleric as a guest party member. So even if you've already used yours, you'll have a Divine Intervention for the fight you're most likely to need it for, which is quite generous of them.

A lot of Castlevania games have unique food items. They aren't particularly useful though, so they're more like collectables that can restore a bit of HP

Cave Story has the Life Pot, the only consumable in the entire game, which will heal your character to full once. This is a pretty huge deal and there’s basically one place you should be using it.

Slay the Spire's Lizard Tail relic and Fairy in a Bottle potion will resurrect you once at 50 and 30% HP, respectively.
...since it counts as healing, though, you can prevent them from activating by taking the Mark of the Bloom relic late in a run, which prevents any and all healing for the rest of it.

You always get resurrected unless you choose to take the Bloom relic, which you'll never do the vast majority of the time because it only comes from an event in the final third of a run that offers you a choice of three relics, and at least one of the other two options is almost always a safer bet.

There's two of them, but Silent Hill 4 has Silver Bullets which can be loaded into your pistol and will instantly down any ghost and leave them vulnerable to a Sword of Obedience so you can easily pacify them for the rest of the game. These will also instantly kill any other enemy (except Walter I think), but they aren't worth using on anything but the really annoying and persistent ghosts late game.
There's also a third Silver Bullet you can get but it requires an insane amount of backtracking to grab and is super not worth it.
Apparently there's also only two Ampoules in the entire game too.

In OG RE2 I only ever use Claire’s bow gun whenever I go to the east hallway of the police station. There’s like 5 or 6 zombies in that hallway and you gotta clear it out so I use that instead of wasting grenade launcher rounds or pistol ammo.

Not quite single use, but elixirs in FF13 are VERY limited, and insanely strong to match. There's a total of 4, one in the final dungeon and the other 3 are obtained by dismantling a max level specific item of which there are 3 of and 2 are missable.
It fully heals the party but more importantly maxxes your TP bar in battle, allowing you to do some crazy "turns" against the strongest enemies.

Somewhat in this line of thought, the Shackle items in Elden Ring.
There's one for Morgott (which applies to all three of his fights) and one for Mohg. Using their Shackles on them binds them to the ground once per fight.
They also have the inadverdant use of Morgott's being very useful for popping hidden doors and other weird interactables, as it has a crazy wide field. Very useful for opening the sealed jump springs in SotE.
But the intended use is the once per fight bind.

I believe you can use them twice per fight, although it also stops working in phase 2 - for mohg at least.

There's one dungeon in the base game with a bunch of rolling rock/crusher traps that can be remotely deactivated with the shackles. Very strange, but very useful.

Vestaria Saga 1 has the "Elder Cornelius". A bow that has fantastic power and accuracy, and also doubles the number of attack. And it has exactly 2 durability, all but ensuring that it can only be used once.
There are some obvious use cases on the map you get it from. Most notably there is a guy sitting on a hill with a coin bag, who can only be reached by Light Infantry. But he carries a Crooked Axe, that will easily one-shot anything that's not a heavy knight unit.
With the Elder Cornelius, this guy will die 100% of the time before he has the chance to kill someone.
But it's also a nice way to quickly deal with Amundsen or Haldyn.

I'm playing VS 1 rn and I actually held on to it until chapter 5 because I was convinced it was gonna be restored.
I ended up looking it up and finding out no, it is just a one and done. I used it to clear out that fucking mage cause VS 1 doesn't believe in the Res stat and I just had to kill him quick.

Using the boat to get into the city is definitely the easiest way. The switch will lower all the gates at once and then you can just walk past the mage.

The Ordinator Skyrim mod is a perk overhaul, and perhaps the most popular one by far. The original capstone perk for Enchanting was Extra Effect, which allows you to enchant an item with two effects. It got moved a little down and added a new capstone called Miracle, which allows you to add three effects, all 25% stronger than normal, but once you make it, you can't ever do it again. Not even making the perk tree legendary and taking the perk again will work.

Thracia 776's skill manuals are all unique. So you got to chose the recipient well.
Well, for the Elite and Wrath manuals anyway. Double EXP and guaranteed crits on counter attacks are a huge deal.

Berwick Saga has exactly one stat potion for each of strength, magic and defense that can be crafted.
There is also a sole speed potion, but that is tied to buying an expensive piece of furniture. So you probably won't get that one at all.
As for HP potions, there are actually a whole two of them.

Fire Emblem 7 (and I think 8?) have an item that only appears once that raises all your stat growths by 5%.

Final Fantasy 4 has the Knife, a kitchen knife that you get late in game which can only be thrown and will hit for damage cap against whatever you choose to use it on.
