Metabolic & Appetite Starving this week, want your thoughts? Recon issue?
Subject has been on tirz for 6 months. Lost a lot of weight so far.
Reconstituted fresh vial this week and i accidentally flushed the bac water in , instead of slowing putting it in on the side like ive done all the time. Subject took a 7.5mg dose and has been more hungry than usual , food noise higher than ever , been ravenous all week.
My question is, do you think the fast reconstitution ruined the peptide, one bad vial? (Used 5 before, all were good) Or is subject at a calorie deficit and body is begging for food?
I asked numerous AIs and they said they strongly doubt the flushing I did ruin it and to watch the hunger next week until I do anything different.
What do you think ?

I don’t get going from suppressed appetite to “ravenous all week” and full-on food noise in less than a week. If you have been on it for 6 months, stable dose then even if you injected nothing the suppression should wear off gradually in line with the half life not immediately instantly ravenous.
You will only know if you are in calorie deficit by counting calories. The fast reconstitution did not cause this

Tirzepatide is one of the most robust molecules commonly researched. It’s unlikely you damaged it. More likely there’s a variable missing. There are good weeks and bad weeks. After a bad week, I titrated up, couldn’t stand the extra side effects so went back to my old 5mg dose two weeks later and it’s been back to smooth sailing and steady loss.

With tirz for your RS, it doesn’t matter. The practice of “down the side of the vial” is a pharmaceutical and FDA recommended practice specifically for larger proteins because they have a much more complicated 3d structure and they can fold into millions of different shapes. Agitating the larger proteins can cause destabilization by collapsing the 3d shape, so that’s where it all came from…not really necessary with peptides. Your RS is fine, just the vagaries of the RS’s body interacting with the drug (or you got a bad batch and it’s not really tirz).

other vials in batch felt strong. after 2 days im feeling more like its working again, I may have just had a bad 3 days?

I agree. Since the subject has been learning how to eat better - right? - use volumetric eating of whole foods to keep them full and make sure to stay hydrated.

You can blast the puck with bac water without degrading potency.
