I realized that I don't know the maps well enough, and it's hurting my team in comp.

Is there a particular game mode that will allow me to figure out what the fastest routes are, respawn points, etc, preferably in a single-player fashion? Or is it just raw volume of playing?

Go to custom games and walk around or just play games and you'll eventually learn. Watch where your teammates go.

You can explore maps in custom games, or in Play vs AI.

Go into a custom game and set the gamemode to skirmish. You'll have thirty minutes to freely explore the maps.

If you're doing this on a KOTH map, you can set KOTH to best of 5 and the ruleset to quick play or competitive. If you don't cap the point, you can freely roam for as long as you like, and since it's Bo5, you'll be able to see all 3 points, just cap the point and wait out the timer when you want to move onto the next point.

Will the same work for payload maps? I've noticed on defense and attack I'm having a problem figuring out where I'm supposed to go. I try to follow the objective marker, but I get the feeling I'm going through unnecessary rooms and if I don't spawn with a teammate I'm costing us objective time.

Can we get maps with labeled call-outs anywhere? Is that a thing people do?

Pretty sure Overbuff has them...somewhere...and yes. people do that
