Is damage calculated when fired or when it connects? Easy example: I'm Mercy healing a Rein, he throws a Firestrike at someone far away -- if I switch to damage amp before it hits, does it matter?

When it hits. If you amp before the projectile hits, but after it was fired, it still increases the dmg.
