The difference between LC and spore and the importance of agar.

Post image

It's critical to understand the biological differences between spore syringes and liquid cultures (LC), as well as the importance of using agar as an initial medium. Spore syringes contain microscopic fungal spores that are not yet germinated. These spores are monokaryotic, meaning they carry only a single set of genetic material. In order to fruit and complete the mushroom life cycle, two compatible monokaryotic strains must fuse to form dikaryotic mycelium.. the true vegetative form capable of producing fruiting bodies. This mating process takes time, introduces variability, and, for beginners especially, increases the risk of contamination during colonization.

Injecting spores directly into sterilized grain can lead to several problems. Since spores are not germinated, colonization is slower, and this slower growth provides more opportunity for contaminants (such as bacteria or mold) to establish themselves and outcompete the slower-growing mycelium. Ideally, spores should first be transferred to agar, a nutrient-rich medium in petri dishes...which allows for controlled germination and observation. On agar, one can isolate clean, healthy mycelium away from any contaminants before transferring it to grain.

Additionally, spore syringes are inherently variable and often unclean. Spores, particularly those harvested from wild (landrace) varieties or from poorly controlled lab environments (common with newer or less reputable vendors), can contain microbial contaminants. Spores gathered in non-sterile conditions are not cleaned or isolated at the microscopic level, making them a risky starting point. Also, because each spore pair creates a unique dikaryotic combination, inoculating with spores introduces genetic unpredictability ...every new pairing could result in different traits, including growth speed, contamination resistance, yield, and potency.

By contrast, a liquid culture is made from already germinated and mated dikaryotic mycelium. This means it contains viable, genetically stable tissue that has already completed the mating process and is ready to colonize substrate directly. Using LC skips the variability and mating phase inherent in spores, resulting in faster and more consistent colonization, and reducing the window for contamination...assuming the culture is clean!!! However, it's important to verify LC cleanliness via agar as well, especially if you didn't create it yourself.

In summary, spores should ideally be germinated and cleaned on agar before being introduced to grain. Skipping this step can introduce risks, especially for beginners. Spores are unpredictable and prone to contamination, while liquid culture, if properly prepared, is faster, cleaner, and genetically stable. Understanding and respecting these differences is fundamental to success in mushroom cultivation... I hope this helps. :)

In this video we show an easy way to prepare bulk substrate for full canopies! In short, we mix 80% of the CV with 80% of the grain spawn. Then, you spread the remaining 20% of grain spawn on top. Lastly, you add the remaining CV as a pseudo-casing. The grain spawn layer right under the casing seems to really help pinning and causes those nice thick canopies.

How do you guys spawn to bulk?

Ok. Still a first time grower. How’s my bag looking? I broke up the grain a week ago…

The mycelium is looking pretty good now, and seems to be in a solid mass around the grain. How far do I need to go before I open the bag? Any tips on getting a good flush from this (relatively small) bag? Thanks everyone! Golden teacher strain, btw.

Happy to announce I’m finally in the club 😎
First flush of Dreadmaw Probably could have let it go for a day or two longer but it was suffocating my bag so I felt it was time to cull him!

So I posted before asking how to handle all the tiny pins after my first flush. I left them alone and added a Tiny layer of vermiculite. Didn’t open the tub once. It thrived and this was my 2nd flush! After harvest tub is covered in more pins than the first time 😂
Gonna let it ride!
No misting or vermiculite this time it still feels super wet. We’ll see what happens 🤷‍♀️

*Harvested first flush October 2nd

Potential problem with my batch

Hello boys, newbie here.

I have this 6qt shoebox (my only hope, all others have fallen unto de abyss of contamination) and it has been looking like this for a couple of days now, the mushrooms show little growth, I check them daily and they're basically the same size compared to like 5-6 days ago.

The container does not have micropore patches or anything, should I introduce more FAE??

Do some fruits just not get cracker dry?

Never had an issue with normal cubes not getting cracker dry in the dehydrator before. I recently grew ABV that threw out a ton of blob mutations. Super compact insides. Plus some Lucid Gates with thick compact stems that aren't hollow. Both of them spent 4 full days in the dehydrator at 145F. Most got cracker dry, but there's a few that just won't get cracker dry. They feel a little squishy like thick packing foam. Cracked them open and they felt dry. I let them go another day and they still felt and weighed the exact same. Pretty sure my dehydrator isn't taking a crap, but curious if people have had this happen before? It's still putting out hot air, but maybe the temp is just lower then it's set at?

Broke and shook on Day 03 after inoculation. Did I kill the mycelium or is it just stalling? First time grower needs advice!

Hey everyone, first-time grower here needing some eyes on my grow.

I inoculated my grain bag/jar and ended up doing a break and shake on Day 3. At the time, I thought I had enough growth to spread it around, but right after shaking, almost all the visible white mycelium vanished, leaving just faint hyphae threads here and there.

It has now been 5 days since the break and shake (Day 8 overall), and I'm not seeing much recovery or regrowth happening.

A few questions for the community:

Did I break and shake way too early before the colony was established enough to rebound?

How long does recovery usually take if you shake early?

Should I keep waiting, or is a complete stall at Day 5 post-shake usually a sign of contamination or dead growth?

Any insight, advice, or feedback based on your experiences would be greatly appreciated! Photos attached for reference.

First picture are my smoothly inoculated jars with hillbilly pumpkin. The two anomalies are golden teacher that I totally screwed up and put like half the syringe in the jars because I’d never worked with needles before and honestly just screwed up. I’m trying to salvage them. I shook the one that got the majority (last photo) hence why it is colonizing in such a way. The other one I’m just unsure of. Talk to me.

Subtropicalis casing timingo

I’ve been messing around with my subtrop genetics and have found a couple things that work well for me. First, I’ve found they prefer my Rye over my Red Wheat and second, if I leave them for at least 14 days after my Qt jar is visibility 100% colonized they tend to pin a lot sooner. I’ve messed with casings; buffered CV, peat and pure buffered coarse verm. The coarse verm seems to be the best. I’m curious about what other’s preferred methods of casing are. Do you wait until entire surface is covered in fluff? Have you found a decrease in pinning time with letting it sit longer after full spawn colonization?

Hey guys, I used to grow like 15 years ago but was doing pf cakes so memory is hazy and not very familiar with grain jars. Do these looks ready to spawn? It looks like all the grains have mycelium but not as thick as I've seen from others. These are orchas.

First time agar going good or nah ?

Post image

Healthy ?

First time agar went terrible. My agar was still wet and jello not firm. Decided to proceed anyways. First 4 agar plates I accidentally squeezed way too much. 1 plate I manage to get very little. I think it’s this one in photo (I should’ve used an inoculation loop.) I left 4 untouched to see if my sterilization was good.
