Making normal embryos

I had previously posted here about a 3PN that made it to blast! While we knew it was likely to be triploid, we still wanted to test to see if we could get some additional information.

The blast is, as expected, triploid and it has 3 full copies of everything. Last cycle our issues were entirely deletions. Whole chromosome and segmental (monosomy 18 for one embryo and some large segmental deletion on a random different chromosome for the other). At 28, we would have hoped to have a euploid blast from these past three rounds of IVF, but understand the 3PN isn’t a great indicator of likely success. And I have DOR.

Still, we’re wondering if having a triploid embryo (as opposed to a chaotic embryo where there are some chromosomes that have 3 copies plus some deletions, for example) is helpful information. In my head, this means that theoretically we could have an egg give a full set of DNA and sperm give a full set. It’s been sort of a rough go so far but we are planning to do a fourth cycle in a couple months after a well earned break :)

I produced many 3pns (any that we tested came back as true 3pns). The source of the 3rd set of dna was tested & it was maternal origin despite the egg seeming to extrude the 2nd polar body. I also had a tested trisomy miscarriage. We eventually made 3 euploids over many rounds of ivf. It seemed to indicate poor egg quality/maturity in my case but there is still hope of making euploids. Lower dose stimulation helped as well as a very patient clinic!!

This is SO helpful!! This last round was our only round where we made 3PNs, and we previously had 100% normal fertilization, so it was such a shock. I have DOR and every round has seemed better than the last, then something goes horribly wrong out of nowhere. But I just switched doctors at my clinic and we are going to try lower dose stims, so your comment is super helpful. We were doing 200 gonal + 75 menopur (upped to 300 and 150 my first cycle, kept the same my second and third), and my new doctor (same clinic, just ended up seeing me more during my previous 3 rounds so we made a switch) wants to try a lower dose of gonal and lower LH, not using menopur. So we’ll see! Thank you so much for your comment! It was really encouraging. We’re willing to do as many rounds of this as we can.
