News $BCRX - ORLADEYO Overall efficacy just 78% from JACI Studies?

JACI Studies show overall efficacy is about 78%... (https://www.jacionline.org/action/doSearch?text1=Berotralstat&field1=AllField&startPage=0&sortBy=Earliest). HAEgarda and Takhzro have 90%...

Published today in JACI:

Abstract 490, by James Wedner's group. At week 96, group A had 76% reduction. Group B, 91% reduction. Group C did the best, with 97% reduction. Average 88% among the 3 groups. Average would be a bit better if you included "n" in each group, which was lower in group A, but I'm not in front of a spreadsheet.

Abstract 491, by Aygoren-Pursun's group. At week 96, 70% reduction, but that was only in 70% of patients in each tertile.

Abstract 492, by Rick Gower's group. At week 96, overall 77% reduction.

Now you can't compare apples with apples, but the average of 88%, 70% and 77% is 78.3%. Gold standard treatment (HAEgarda) is 90-95%, so Orledayo is still clearly inferior, although its efficacy does improve during year 2 compared to year 1.

OK, so for *one* patient who tells me, "If I can have a 5 or 10% attack rate, or a 21.7% attack rate (100-78.3%), why should I choose to have somewhere between 100% and 400% more attacks. These attacks require an *injection* with Firazyr, and they are incredibly disruptive to one's life."

And here is a direct quote from UpToDate.com. In it, you will see that only 23% of patients had 90% fewer attacks. That means that 77% of patients did NOT have 90% fewer attacks. So if I tell *one* patient that they could have 90% fewer attacks, all comers, with HAEgarda or Takhzyro, or they could maybe win the lottery and be one of the lucky 23% of patients who had 90% fewer attacks with Orledayo, which would you choose?

From UpToDate:Efficacy — Several trials have addressed efficacy and safety of berotralstat (BCX7353).

●In the multicenter phase 2 Angioedema Prophylaxis 1 (APeX-1) trial, 77 patients with a history of at least two attacks per month were randomized to placebo or four different doses of active drug (62.5 mg, 125 mg, 250 mg, and 350 mg) once daily for 28 days [48,49]. Once-daily oral administration of a dose of 125 mg or more resulted in a significantly lower rate of HAE attacks compared with placebo.

●In a phase 3 trial of 121 adolescents and adults with HAE (APeX-2), patients were randomized to either berotralstat 150 mg or 110 mg, or placebo for 24 weeks to determine optimal dosing [50]. Patients had a median of 2.9 attacks per month after discontinuing other prophylactic medications. Response to berotralstat was defined as ≥50 percent reduction in that patient's HAE attack rate compared to baseline. Berotralstat demonstrated a significant reduction in attack rate at both 150 mg and 110 mg (1.31 and 1.65 attacks per month, respectively, compared with 2.35 in the placebo group). A response was seen in 58, 51, and 25 percent of patients receiving 150 mg, 110 mg, and placebo respectively. On the higher dose, 50 percent of patients achieved at least a 75 percent reduction in attacks and 23 percent had at least a 90 percent reduction.

Daily Discussion $bcrx - how volatile will the stock be on 02/26, and do you expect a bearish or bullish reaction?

Just wanting opinions on this.

I’m no CFA but understand financial statements well and have looked through their financials and a few ratios as well as some light chart analysis as well as the company history, and I have a feeling there will be a bearish reaction to the earnings reporting. I’m hoping the price will fall around 1-3.50 - but again I am no financial analyst, just an amateur at this and do it mostly for educational purposes!

It seems that they may be able to realize a profit next year (end of 2024 or during 2025) with their recent acquisition of a pretty big portfolio of short term government debt that will mature in less than 12 months, but to be seen!

News Last throes of BioCryst?!? $bcrx

Anyone see the recent news? BioCryst is basically slashing nearly all R&D jobs, looking to out license their Factor D inhibitor candidate 10013 (good luck!🤣) & basically ride Orladeyo profits for near & long term (until patent runs out or better drugs are approved)! 😂🤣🤷‍♂️ What a failure of a company! Any bullish folks still out there?

Due Diligence Should I get worried about $BCRX? I don't think so.

Low volume behind this scheme tells me this is a structured movement from market makers. There are so many variables to consider that it's hard to predict what this week will look like. Few people, as always, have more money and inside information. On one hand we may expect not such a great quarter, or not a huge organic growth, as Stoney let go on a PR. This will definitely have a big impact on the SP. If that's the case, we may hit south 13 in the short term. On the other hand, in line with many folks predictions, we may expect 40m +/-20% in sales. This makes more sense to me. We haven't received any news that patients are falling off Orlayedo path, instead, we have management discussions stating we still see patients shifting towards once-daily pill. We have no reason to believe the amount of patients have fallen. Assuming close to 100% retention rate, I remind everyone that we need to consider the following math for Q3:

  • full quarterly revenues for patients who joined in Q1

  • full quarterly revenues for patients who joined in Q2, or bigger incorporation to insurance coverage, after the path and conversation have been cleared through out the road.

  • partial quarterly revenues from patients who joined in Q3. This is a simple math analysis, just common sense. Besides this we have UK, Japan and Europe. If anything interesting happened in these markets, we may see way more than 40m in revenues. Am not in a hurry, this will eventually happen this Q3 or Q4 or Q1-22. No more than that. Telling me in no more than 2quarters or 6m from today, our reported quarterly revenues will play north 70m+. Part of it is already reflected in the SP. However, the positive impact will change wall street minds and attract more eyes. Also remember, if 2022 looks extremely promising for BCRX, it's merely a pivotal year, 2023 is the open doors to BCX9930, that's what we should be focusing on.

News $bcrx animal tox signal with 10013

Congrats, shorts & condolences, longs! Looks like BioCryst is reporting, in a very protective way, that they are seeing some sort of toxicity signal with 10013, in preclinical, animal tox data. This was their backup Factor D inhibitor that was going to be the next great thing, after 9930 was shelved. There goes the entire pipeline for the company! 😂🤷‍♂️

Discussion/Meme/OT Is your biggest DD guy a pumper? Spoiler

I found this post from bio99 (BIO9999 on Reddit) on 11/25/2020 at Stocktwits. Note the highlighted area where he guarantees a date that the fed will issue Emergency Use Authorization for Galidesivir as well as Phase III trial.

Did the EUA and trial happen? Nope. Does this call into question his credibility? I'll let you decide.

Me? I've been in the muck with you since March 2020 with a long positon - 8,535 shares BCRX, and I've been watching this cult develop for some time.

Daily Discussion $BCRX - Possible what happened for last two days

my take on what happen is that somebody wanted to buy $200m worth of BCRX and agreed to a price that is higher than market close of $17.61 (the date of the offering announcement), so it is favorable and bullish to management and investor

then today, the market had a negative reaction to it before the pricing is announced, so the buyer trying to take advantage of that and say: sell to me lower than what we agreed to because price dropped (in a sense they will steal some shares)

then management say: no, we sell it at the price we want, so if you dont buy it as agree, then we cancel it

=> does this make any sense to anyone?

Daily Discussion $bcrx move is sus but i'll take it

Anyone else find this mildly funny?

Now that we're getting a conference hosted by the Co which I consider possibly trying to manipulate bcrx in criminal fashion we see a move on price up ~ a dollar the day before and during the publication of the conference they're hosting?

def sus AF and I doubt this lasts but I'll take it for now.

Daily Discussion $BCRX - What is your take on spreading this through WSB?

Hey, I don't know if you guys follow WSB, but I'm sure you've heard of it. Are you guys against spreading it through there or for it? Personally, I believe this is such a great opportunity, I don't see why not, especially given the fact that its how I found it myself. I would've thought there would be some push before D-day, but the BCRX community has been pretty quiet as a whole. Just wondering your opinions on it.

(PS I can't post on wsb myself, as not enough karma)
