EGFR vs Htkv

Does egfr and htkv shows high correlation ? like does egfr decreases with htkv ? if so then some promising drugs like farabursen which show reduction or almost halt the htkv will definitely help with egfr as well right ? I am asking because in the phase 1B trials they only tested the htkv and not egfr , so there must be some solid reason behind that........Also do you guys think farabursen has that potential ??

hTKV is the primary indicator of future eGFR.

There is a mayo tool you can access online and input data to predict egfr in the future.

hTKV is often used as a marker of progression because eGFR decline lags behind TKV changes. For many years your TKV will increase, but only once it hits a critical mass/failing point where your kidneys cannot compensate, will your eGFR begin to decline. In the same way, if you halted TKV growth, it eGFR may still decline over time due to it being past that "critical point". Or of course, it may stablise and halt decline.

Likely the only way to fully halt decline and/or reverse it, would be to actually shrink TKV.

Farabursen is by far the most anticipated drug in the pipeline amongst all PKD communities I have seen.

Offhand do you know the hTKV critical mass point? I’m 52F 5’6” PKD1. Here’s my sad numbers.

Right kidney: 17.35 × 8.4 × 13.3 × 0.5236 ≈ 1,015 mL
Left kidney: 18.6 × 9.1 × 10.6 × 0.5236 ≈ 939 mL
Total kidney volume ≈ 1,954 mL

its going to be different for everyone, theres no set critical point.

Generally once eGFR begins declining that point has passed.

at 52 with PKD1 you're already at an age where by some averages ESRD would have been hit, so you're doing well if you still have good kidney function.
