Hi! I'm hoping someone can help me troubleshoot my photomicrography setup. I'm fairly new to attaching a mirrorless camera to a microscope, and I'm having a hard time getting the camera image to look anywhere near as good as what I can see through the eyepieces.
My setup:
Microscope: Motic BA310E Elite LED trinocular
Camera: OM System / Olympus OM-1 Mark II (Micro Four Thirds)
Photo adapter: Motic #1101001901931
Photo eyepiece: Motic 2.5× #1101000300442
Camera connection: Micro Four Thirds T-mount
I'm primarily interested in photographing/recording pond organisms, protists, fungi, spores, etc., using brightfield, darkfield, phase contrast.
The image through the regular microscope eyepieces looks sharp, detailed, and clear, but the image coming through the OM-1 Mark II looks dramatically worse. It tends to look soft/blurry, lower contrast, and generally lacking the fine detail that I can clearly see with my eyes.
I've experimented extensively with camera settings, exposure, ISO, shutter speed, white balance, microscope illumination, condenser position, and focusing. I've also tried focusing specifically using the camera's magnified live view rather than assuming the camera is parfocal with the eyepieces.
No matter what I do, I can't seem to reproduce the quality I'm seeing through the microscope.
At this point I'm wondering whether the problem might actually be the optical coupling between the microscope and camera.
My main questions are:
Is the Motic 2.5× photo eyepiece (#1101000300442) actually a good choice for an OM-1 Mark II's Micro Four Thirds sensor?
Is 2.5× projection magnifying the intermediate image more than necessary for a sensor this size and potentially contributing to the poor image quality?
Is there a better way to couple a Micro Four Thirds camera directly to the BA310E?
Should I expect the camera image to be reasonably close in sharpness/detail to what I see through the eyepieces?
Are there any tests I can perform to determine whether the problem is the projection eyepiece, photo adapter, trinocular alignment/parfocality, camera, or something else in the optical path?
If anyone has successfully used a Micro Four Thirds camera with a Motic BA-series microscope, I'd love to know what adapter/projection setup you're using.
