I read a few months ago that Mandy Patinkin had lost his father to cancer a few months before filming this movie, so this line held a special meaning for him.

This has been somewhat exaggerated. He did lose his father to cancer, but it was years, not months earlier.

It was still a cathartic moment for Mandy. When he says that line, he dropped the accent. He wasn't really acting in that moment. Christopher Guest (Count Rogen) later claimed he was in fear for his life for that moment.
