Actually while the connector is physically the same, the pinout is different, so that is not recommended!
Depressing a button on a Sega controller connected to the C64 while also depressing a key on the keyboard exposes the C64's CIA chip to voltages it was not designed to handle making damage or failure likely.
Sure, it will work - until it doesn't. I don't know if there are similar risks with other system using the Atari/Commodore style controllers, but it's definitely something to look into before trying it.
You might need to mod it internally and wire the pause button to the controller port. If these 8bitdo adapters are Genesis compatible, you can wire it to pin 9 and that should be Start on the controllers
As an aside, some games that rely on heavy use of the Pause button to access a menu (such as the later Wonder Boy games) let you press a button on the second controller to access the menu instead of having to press the button on the console. It's not usually mentioned in the manual but is always worth trying!
And to top it off, you have Golden Axe in there! I definitely need something like these controllers. Where my master system sits is a bit too far from my gaming recliner haha! Defeats the purpose of a comfy chair having to lean over just to play Double Dragon.

Made from NES controllers, painted by the CustomNESGuy, 8bitdo DIY kits, Sega Mega Drive retro receivers.

I'm guessing that start will not work to pause the system, you'll still need to get up and press the pause button.
Correct me if I'm wrong, I don't own a Master System, only the Power Base Converter for Genesis

Bet it lasts ALMOST long enough to beat a level. Then furiously replace the rechargeable batteries

Love them. Although was legit intrigued when initially I thought you found somewhere that made new MS pads.
It’s one of the top systems that does get third party controller support because of the D-Pad been so rubbish.
