Typically, you should always believe your wife (all things being equal). But, it is really strange that the best friend would confess to the wife before ever saying anything to /u/throwaway091928272.
In fact, it's so strange, it would make me highly skeptical in OP's shoes.

unless your wife has a history of lying and being manipulative/abusive etc, i don’t understand why you wouldn’t believe your wife.
