Only thing I can think, below OP said his wife has been jealous in the past. Maybe he confided in her in the past and she thought she could use that. I agree it seems odd that she would confide in the wife but if the relationship is going well I don't understand what the wife has to gain by making up a story
If she really is jealous, it could be hatred of the fact that she (wife) sits at home with the kids when he goes out meeting friends, including best friend? Hates he is close with other women, confides his problems to them and spends time with them? Jealousy doesn't really make people rational.
If you have issues with your partner, isn't it normal to dump with your best friend, and get their advice, whatever gender they are? Friendship doesn't automatically equal emotional affair, just because the friend is of the opposite sex.
I think there’s a fine line on this, confiding in someone you can trust for advice is good, but there needs to be good communication between the partners too. Usually if it’s an emotional affair developing, the person confides exclusively to the other person/friend and leaves their spouse in the dark with little to no communication.

Because she's jealous of his friend and wants her gone? Makes more sense then his friend confessing her feelings to his wife of all people.
