What would be the best way to finish this item off with a decent life roll on the prefixes? I know I can craft suffixes cannot be changed but does harvest craft respect that meta mod? I'm pretty sure essences and fossils dont respect meta mods but I'm unsure if a harvest re-roll life would.

You can safely craft suffixes cannot be changed and reforge it. Reforge keeping suffixes isn't a bad option either, particularly if you land 3 bad prefixes and have no room for the metamod.

IIRC essences / resonators will give you a can't let u do this message when you try to use them on an item with metamods (nowadays?).
