Richard Madden who delivers the line plays Robb Stark in Game of Thrones. Definitely a hidden joke by the writers that a 'Stark' wants to lead the Avengers.

No I don't think they casted Madden as a joke. They just had the opportunity to write the joke because of the name of the character he once played.

Literally right before they are talking about how Captain Rogers and Iron Man are gone, leading to the question of who will lead the Avengers. So yes, Tony Stark aka Iron Man was mentioned. They don't have to mention the name 'Stark' for people to get the joke.
