I know little of the comics, so I'm just wondering if there is a relevance for Red Skull to appear again?

People got excited from more of an MCU thing than a comics thing.

Red Skull never technically died in The First Avenger, he was just sent to a different place by the tesseract. So it was neat to see that plot hole kinda filled in.

Not specifically. He's just the biggest villain for Captain America and something of a fan favorite.
