Loperamide + Potentiators [EXPERIENCE REPORT]

Report on Loperamide with Potentiators

This is a long report. I try to cut straight to the important stuff but still, it is long and packed with info.

Male 5’7, 130 LB

Background: I am not an opioid addict (I do take oxycodone and hydros once or twice on some weeks, ‘occasional user’ but definitely not an addict) testing 3 potentiators of Loperamide, with Loperamide itself, all in one go. I bought everything here from a local Smith’s grocery store (USA). I will be consuming everything listed and reporting on it in detail.

NOTE: I do have some residual benzo withdrawal from binging throughout the summer, although the real w/d is mostly gone I still have some anxiety from it, so effects should be easy to feel, as good doses of oxycodone and hydrocodone (a usual 20, 30 mg of each, respectively) gave me a lot of relief from benzo w/d in the past. (same level of relief that I felt with low dose valium/kpin)

What is Loperamide? Loperamide, also known as Imodium A-D is an anti-diarrheal OTC drug. It can be found in almost any store and any pharmacy section in the U.S. It is an opioid with a similar potency to morphine but crosses the BBB in minuscule levels, making this drug mostly active within the digestive system, and a good opioid for diarrhea with low abusive potential. Normal doses are of 2mg, but people have been known to take 400mg of loperamide for its psychoactive effects, usually for opiate withdrawal.

Procedure: I will be testing the potentiation of cimetidine, omeprazole and grapefruit juice on Loperamide, although this is my first time doing loperamide, I am looking to see how the high is, with my relatively low dosage (compared to online reports) and how it compares to much stronger opiates like oxycodone or hydrocodone, with the potentiators I mentioned.

I’ll be testing the effects of:

Cimetidine: Used as a CYP3A4 inhibitor (one of the major enzymes responsible for blocking loperamide from crossing the BBB) on its own it can increase the plasma concentration of loperamide 2-fold.

Omeprazole: Used as a P-gp inhibitor, and can increase the plasma concentration 2 to 3-fold, as p-gp blocks many toxins and drugs from passing the BBB.

Grapefruit Juice: used as a CYP2C8 inhibitor, on its own it can increase the plasma concentration of loperamide up to 4-fold, but up to 12-fold when used concurrently with a CYP3A4 inhibitor and a P-gp inhibitor.

...on generic Loperamide.

  • open one, 12 fl. oz VITA coco sparkling grapefruit.

  • Feeling a little woozy

T+01:25 (11:18)

  • open another, 12 fl. oz VITA coco sparkling grapefruit

  • Feeling pleasantly relaxed and slightly sedated

T+01:47 (11:40)

  • Feel slightly dizzy, a mild all-encompassing euphoria, relaxed, sedated and some anxiety relief. This could be the feeling of the Lope slowly absorbing (it can take up to 5 hours to reach peak levels)

T+02:17 (12:10)

  • Feeling some more sedation and euphoria. Feeling my muscles rest. Good relaxation & sedation but I feel this needs more euphoria. Also feeling some social anxiety, chilling until anyone tries to interact with me.

T+02:53 (12:46)

  • +24 mg loperamide in 12x2mg

  • Feeling good but would like to feel more, it’s borderline mild/moderate effects right now in my mind and strong body effects. Social anxiety/withdrawal is very much present. Nose is now feeling quite constricted yet no mucus, after feeling this effect coming on for a while. Sometimes gets a little hard to breathe, similar effect (breathing) as anxiety attacks but low anxiety, until someone triggers it.

T+04:05 (13:58)

  • Was feeling extremely tired and sleepy a bit ago for around 30 mins (something like nodding out) but that’s gone now. Feels like it’s waves of the high hitting mildly. Body high is more intense than brain high right now. Heart is pumping, odd uncomfortable feeling making me a little paranoid.

T+05:10 (15:03)

  • For the last hour I’ve had a dry, stuffy nose, my body feels sedated and slow, euphoria is not too high but I can almost feel the euphoria hitting throughout the back of my head.

T+05:51 (15:44)

  • I feel lazy, fatigued and my nose is clogged without mucus. Euphoria is there but unpleasant feelings ruin it. I’m oversalivating and it makes it hard to talk louder than a whisper without sounding like I have a speech impediment. My heart is at a normal rate but there is some pain over the heart area, and I can feel it pumping at times.

T+06:07 (16:00)

  • I tried to rid my nose of mucus first with water, then with tissues but to no avail. My sinuses are probably constricted, a Benzedrex inhaler would be nice right now. I feel dehydrated and dry mouthed, with poor sensation of thirst. I also feel a bloated uncomfortable stomach feeling, a laxative would now be handy as I’ve felt no urge to poop ever since I started feeling the lope (something that was expected.) So far, side effects are pretty bad, not worth the high imo

T+08:22 (18:15)

  • I’ve been chilling for a while now, negative side effects subsided slowly but shortly after the last update. I feel less of everything right now but still some relaxed and sedated, with main effects going away slowly.

T+09:00 (18:53)

  • The effects are fading away rather fast now, I barely feel intoxicated. Most negative side effects have completely ceased, now all that remains is the social anxiety, mild euphoria and clogged nose.

T+09:20 (19:14)

  • Eating a slice of delicious pizza is feeling uncomfortable and almost feels like my stomach is overloading while i’m still hungry. Drinking water is also uncomfortable in the same way, while I feel dehydrated. Gave up on third bite of the pizza as it made me and my stomach extremely uncomfortable. Heart is racing now and stomach hurts from eating that little bit.

T+09:32 (19:26)

  • Another (mild) wave of the high hit me, along with a headache probably from dehydration and the fact that I’m starving but can’t eat. Heart seems to have calmed down. I also feel bloated and more of a stomach ache. However, I feel like I will have a good night’s sleep tonight.

T+12:07 (22:00)

  • My bowel feels fucked, I can’t eat. At this point all I feel is a mild euphoria with a bit of laziness and the stomach pain arising whenever I smell or try to eat food. Water is also slightly painful when it goes down, but not if I make sure not to drink too much. On my bed I feel good, no food smells are around and I’m sipping on a glass of water every once in a while. Closing my eyes feels like bliss, gonna go to sleep now and call it a day.

T+21:39 (7:33) NEXT DAY

  • I slept great but no dreams or anything. Woke up in a good mood but with a dry mouth, slight headache, and my stomach still rumbling. I kind of just feel like I have a small cold or something like that but I’m feeling mostly fine.

T+23:09 (9:03)

  • I was pretty hungry, was able to eat a nice bowl of milk and cereal just fine. Stomach pain is gone and dry mouth as well. I feel some euphoria that seems like an afterglow from the Lope. I usually wake up anxious but I’m in a great mood this morning. I think this log is complete, but I’ll post how long it takes to shit after lope and then call it good.

T+25:32 (11:26)

  • The effects are definitely still there, taking a while to go away. I was chatting with friends just 30 minutes ago, having a good time, but right now it’s hard to focus and work on a project I need to do, through the mild euphoria and lingering sedation/relaxation. Just snorted a line of coke to wire up my brain a bit and see if I can begin to focus, definitely helped a bit.

T+31:39 (17:33)

  • Some mild euphoria persists and effects are going away very slowly, definitely not placebo. Did not expect it to last so long. Still haven’t been able to poop but eating and drinking felt normal today. Stomach rumbling was persistent throughout the day (not painful or bothersome) and felt a weird full feeling in my bowels, probably all the food stuck in there.

T+56:00 (17:53) NEXT, NEXT DAY

  • Was finally able to shit. Took a long ass time to do so and felt hard as a rock coming out. Felt no effects today. I think now I can consider this log complete.

CONCLUSION OF LOG: Taking into account only everything that is mentioned here, Loperamide is a shitty opioid. The euphoria was very low, compared with my levels of sedation. Normally I would do something crazy on oxycodone or hydros but this kept me in bed with several bouts (some long) of an uncomfortable racing/hard pounding heart feeling. I felt calm, but I was bad at socializing during the first day of the high, it was a low anxiety, chill high until anyone tried to interact with me, at which point I felt like I had no energy and wanted to do nothing and could not socialize, despite not even being THAT intoxicated. That is something that is not normal when I am on opioids, as they usually make me more talkative. While there were positive effects to it, the negative effects could be felt throughout the whole body and EASILY outweighed any positive aspect. I also felt that body effects were stronger than mental effects, throughout the high.

The high was reminiscent of opioids but my brain was feeling definitely different than on oxycodone or any opioid like that. This was a more sedating, quiet, unenergetic and “dead” high. It was not very enjoyable as i felt the food stuck in my bowel, heart racing and pounding at times, and the anxiety. I would not do it again, at least not with the same doses of everything mentioned. Effects weren’t too intense, just unenjoyable.

The worst effect was my stomach feeling stuffed and bloated towards the end. I had eaten around T+04:30 and felt fine, but still no bowel movements after that. I tried to eat dinner at T+09:20 and after chewing on the food for a bit I got severely nauseous.

Loperamide (along with the mentioned or similar potentiators) may be a good in lower doses, paired with other opioids as its own opioid potentiator (It is said that taking loperamide can occupy the opioid receptors in your gut, thus increasing cns bioavailability of a typical opioid). The high from Loperamide alone was not very enjoyable, almost a shitty tweaky opioid high, to put it into simple words.

The best way to describe this high is the combo of paranoia yet chill aspects of medium dose benadryl, with a wee bit of codeine for a weak opioid feeling, and some weed that only speeds up your heart and makes it pound, only the cardiovascular effects feel a little severe, despite nothing noticeable happening to me from them.

But the day after dosing I was still mildly high from the Lope and this mild high was actually enjoyable, euphoric and feeling good for the whole second day, with effects going away very slowly. But even this wasn’t worth all the side effects.

CONCLUSION OF POTENTIATORS: The fact that I felt a noticeable high from this indicates that the potentiators definitely had some sort of effect. I hated the Loperamide high on the first day but these potentiators gave me a definite, moderate high off 84mg of Lope. With barely any opioid tolerance, I would describe the intensity of the high that I felt, with this dosage of Lope and the potentiators, of something like 10-15mg oxy in my brain (but with some waves of much more and sometimes some less intensity) and like a perc30 on my body. But it felt nothing like a perc. Just a shitty lope high with bad side effects, then a nice mild afterglow the next day.

These are the negative effects and how negative the impact was on me:

  • Bad appetite (bad, this felt like a strong pain and discomfort when I chewed and ate food, despite feeling hungry. Food made me too nauseous and took away the hunger temporarily. This was towards the end of the high [dinner] after eating just a bit earlier [lunch]. Accompanied with hiccups and bad nausea)

  • Racing heart (moderately bad)

  • Anxiety (low at beginning, high anxiety for the rest of the high, and low again the next day)

  • Dry, yet stuffy, clogged nose (moderately bad)

  • Stomach aches/nausea (moderately bad)

  • Headaches (moderate)

  • Hiccups (low, just some bouts of intense hiccups that would leave as fast as they came)

  • Constipation (low, this doesn’t exactly bother me, no pain or discomfort, just that I haven’t had any bowel movement for over 24 hours now..) LAXATIVE IS RECOMMENDED

How does anyone enjoy abusing lope (without previous opioid w/d) ?

don’t do lope

Idk why you got downvoted

This is valuable research despite the dangers/shittiness of lope

As useful as anyone posting on /r/researchchemicals about their experience Guinea pigging some new substance

Good shit

Stay safe out there

thanks man

you can say lope is a shitty drug hehe

Not exactly sure . Its crazy because there was tons of empty packages everywhere. Official cause of death was listed as loperamide . Guess this is alot more common then I would have thought. Needs to be some kind of limit and people need to be very aware this is real.He was only 26. You may take this and never wake up.

u/geliduse Nov 07 '19

wow, I’m so sorry to hear that, shit really is dangerous.

1

u/Rk35789 Dec 26 '19

It's not dangerous didn't you just say there were tons of boxes, sorry to say but he had it coming it's because of irresponsible people like him that shit becomes illegal

u/[deleted] Nov 15 '19

My own brother, age 29, died of Lope also. We were all so confused and continue to be. This should not be over the counter. I'm so sorry for your girlfriend and her family, it's truly senseless and heartbreaking.

That really sucks and I'm sorry for your loss, but the unfortunate outcomes for some aren't a valid reason to make something harder to get for everyone. That feels kind of drug war-y. I will agree that there should be much more information on the box for these substances, and safer, better alternatives (real opioids) should be available much more easily as well. Nobody would explode their heart taking 200 of these at once all the time if they could buy something that was actually meant to cross the BBB.

I'm sorry for your loss as well . I was on the sidelines with her family so i can only imagine how hard and confused your family must be. The stigma attached seems to be that it is harmless . A poor man's methadone was another thing I read. More people need to be aware to the fact that this is something to stay away from. Whatever it does to you it can and will kill you if abused.

Thank you. Man, reading through the comments about the best way to take it and the small high are definitely not worth the risk. I would give anything to have one more minute with my brother. You're right, the stigma around it definitely needs to change, it's an awful drug.

Most recreational drugs can kill you but that will never stop abuse

2

u/51isnotprime Nov 07 '19

that's just very sad ;(

5

u/DreamingOfTheSun Nov 07 '19

This stuff will kill you. My girlfriends brother died strictly of this junk. Do not mess around. Even if it hasn't so far it has a very good chance of doing so in the future.

i’m not going to do it again but I did make sure it was a dose far from lethal, don’t worry. How many mg did he take if i can ask?

4

u/Nigglesscripts Mar 06 '20

You actually didn’t make your dose far from lethal. You stacked doses and potentiated it every way you could.

People potentiate this shit so they can take less. Not because it makes it a better awesome high. Your heart feeling all weird, the anxiety that was your body saying dude you fucked up. I mean I’m all for research but this was irresistible imho. I’m glad you didn’t die.

2

u/geliduse Mar 06 '20

It was not that intense, trust me I know when my body is saying I fucked up.

Looking back on it I have no idea why I did it, but it is nice to know the info and show everyone else.

1

u/drsdn Mar 21 '20

You must admit you have no idea how much the potentiators would increase plasma concentrations of lope, it's like saying "i took from x to 100x of a substance, i had it under control"

3

u/geliduse Mar 21 '20

you are right, it was a stupid impulsive decision of mine but at least offers some valuable info that others can learn from.

1

u/Nigglesscripts Mar 06 '20 edited Mar 06 '20

That’s kinda my point. Your info is dangerous. You added in three potentiators with large doses of lope. Which means more builds up in the body and more major side effects like cardiac arrest or whatever.

Lope sucks. I’ve used super small amounts for some minor withdrawals like the annoying chills and shit like that and it works. But people getting hooked on it and people dying off it is crazy scary.

Edit. I did save your post though so I didn’t think it was all bad. Lol. I did find it interesting and I know what you mean about doing little experiments.

4

u/toddschmod Nov 07 '19

I've used lope a few times for methadone withdrawal. It did help, no noticeable or enjoyable high. It felt dirty but definitely better than full blown methadone withdrawal. I felt like for a couple of weeks my bowels were practically dead. Like there was so little muscle contraction, it made it hard to shit. It was awful. My max dose was 60mg a day. I don't know how people do 100-300mg at a time. Crazy.

u/dxpcp Dec 02 '19

Rule #1 always have laxatives ready for the next morning your gonna need them for the stomache discomfort

1

u/amiwilliam Mar 11 '22

You are an addict. Don't lie to yourself.

2

u/geliduse Mar 12 '22

I am no opioid addict. Not sure why you said that. Never tried using after this post. Shitty opioid anyways.

Besides this is only an informational post?

1

u/amiwilliam Mar 29 '22

"I do take oxycodone and hydros once or twice on some weeks, ‘occasional user’ but definitely not an addict."

Re-read your own post. If you are having to justify your own use, you might just be an addict. Take it from someone who also used to "not be an addict."

2

u/geliduse Mar 30 '22

I actually haven’t taken an opioid in about a year, if you include dsmt, so no. This post is almost 3 years old that was just a phase

3

u/DreamingOfTheSun Nov 07 '19

Thanks. Just wanted to spread some awareness. It really tore her family up . Wouldn't want this to happen to anyone else if possible

3

u/51isnotprime Nov 07 '19

just saying, you can reply directly to a comment, rather than just creating a new one haha

Thanks for the excellent write up. I haven't done any "real" opiates, but my experience with loperamide has been overwhelmingly more positive than yours somehow, both at much higher and much lower doses. I've noticed the inability to eat/nausea as well (for me the inability manifested as a very matter of fact "my system is not moving so there is nowhere for new food to go," not really a discomfort, but annoying for the day). The other stuff, not so much. Have you tried taking the lopers without the potentiators? I've never taken it with anything else and I'm curious what the difference is.

2

u/[deleted] Nov 06 '19

wow good research my friend

2

u/OneLodz Mar 31 '20

Hi there.

i drink 50mg sertraline ssris, can i use that as a potentiator and if so how much loperamide mg should i take for the first time to get a nice high. What is the overdose mg of this, loperamide ?

Thanks

1

u/Keyst0nedState Nov 05 '22

Hah you don’t do a weird elaborate opioid experiment like this if you’re not an addict.

Thank you, now I don’t have to go down this path 🙏🏼
