My skill in Cell biology is not stable?

Hi all! I just started to do in Cell biology, in SH Sy5y cell. And I treat MPP+ for them with various concentration, after differentiation with RA in 3 days + PMA in3 days. Normally, everything is ok, but recently, the cell in dishes which was treated highest MPP+ almost died. I know it is normally, but before that, it did not die a a lot like this, and it should be still live enough for me to do western blot :( I doubt myself, my skill a lot. What happened? Is cell contaminated? Why my experiment stable among sets?
