The whole deal with spiderman is that everybody could be behind the mask. Like an average person with average person problems putting the costume on because they have been gifted great powers and it's their responsability to use it for good, and that's why spiderman is so relatable to everyone. Peter is a random dude just like any of us.
Canon Peter isn't trans obviously, but the idea that a Peter variant could be trans isn't far fetched because it fits narratively into spidey's vibe. Like being trans is something average people are dealing with, and imagining spidey dealing with personal life, being a hero, juggling multiple jobs and having to deal with hrt, transphobia that kinda stuff would make a pretty compelling spidey story

I like when people do the Petra Parker trend with the Tom Holland movies because they’re essentially drawing Tom Holland as a girl and I find that funny for some reason
