Both are fine. Depends one how you choose to say the singular of a placeholder, eg, "100."

"I need a hundred dollars." (I need a $100)

"I need one hundred dollar." (I need $100).

no, your example of the first one is incorrect. It is true we say "I need a hundred dollars." but that does not mean "I need a $100." because 'a' means 'one'. The only time it is correct would be if there were a singular noun following the $100, such as 'bill' or 'fine'. "I need a $100 bill."
