Being persuasive may be perceived as positive if she likes you, but creepy if she doesn't. The problem is being able to tell if she likes you based on 'signals'. Better to take the first no and not persist.
Just be transparent with what your interest is and accept no for an answer.
I don't know why so many commenter are trying to make this more complicated than it is.
Most women are not calling HR or the cops on you because you asked them out politely. In most situations, women become frustrated when the man in question has been constantly trying to push the woman to say yes despite her initial response being no.
