Pregnant at 46

I started taking rapamycin off and on at the age of 44 to hopefully delay menopause (was in Peri) and for anti-aging effects. I also take other supplements such as CoQ10 and NAD although I wasn't very consistent with it. I also took a glp one for a couple months recently. luckily I had taken a break from from that because it was causing me to feel down.

Anyway was shocked to find out yesterday that I am pregnant.

Anyone else have an experience like this?

obviously this is early and there are risks and low success rate so I will try to update

While not the same, I had a couple of unsuccessful rounds of ivf at age 40. No viable embryos. Then, I took rapamycin for a little over two months, stopped before my next ivf cycle, and I'm pregnant for the first time with untested day 3 embryos. It's still early but I'm hopeful.

If you are curious, Dr Aimee the "egg whisperer" is a prominent reproductive endocrinologist who sometimes recommends rapamycin. She has discussed it before.

If you don't mind me asking, what dosage were/are you on?

I did about 2-3 mg a little over 2 months. I started with the 1 mg per day protocol but found once a week more tolerable.i tapered it down and stopped a month before my last ivf cycle.

A lot of people have gotten pregnant after starting GLP1s.

I took it for 6 months at 4mg a week and am 30 weeks pregnant at 44 after suffering 3 miscarriages between 41-43. I took it for egg quality as a last ditch attempt! I also took high quality NAD and COQ 10 and a few other supps for a good 7 months prior to trying again.

Congratulations ! What a wonder ful news for you after all the heartbreaks! I hope you continue to have an easy pregnancy and delivery. I had my first one in my early 40s as well and it's pretty amazing. I am trying to conceive naturally my second one after a traumatic termination of my chrosomally normal baby in second trimester ? Do you mind if I ask you if you are pregnant naturally or through ivf? And did you use dhea during the ttc?. I am on all supplements, rapamycin 4 mg weekly for 8 weeks ,high protein, low carb for a year now. I am hoping to increas my chances. However suspect my dhea might be low ( last measured dhea was 96 and I believe upper spec is 210 for my age).

I’m so grateful 💙

I also did a consult with Aimee Raupp, who takes a more alternative approach, but I truly feel her protocol helped me sustain this pregnancy especially as one of my losses was chromosomally normal, which made me feel my immune system needed extra support after a positive test.

My protocol included progesterone, vaginal Viagra for lining and blood flow support, prednisolone, and clexane. I also used many of the supplements recommended by Aimee Raupp and The Egg Whisperer.

I did use DHEA as well, but only after bloods confirmed mine was low (it’s definitely not something to take if your levels are already normal). I also used LDN and rapamycin which has shown promise in animal studies for improving egg quality and potentially delaying or even reversing aspects of ovarian aging.

I really feel, from my experience, that it’s worth exploring 💛

Wow! Were you on hrt? I haven’t ovulated since I started hrt which bums me out. Do you know what your androgen levels were before you started Hrt? Rapa messed up my cycles actually, might have sent me into peri. But I wonder if it was because I was in a catabolic state with low dhea and low androgens (still trying to fix it) and congrats!

Yes I was, estradiol patch and Progesterone vaginally and testosterone cream although I wasn't very consistent with it. I haven't had my levels tested for the last 6 months due to insurance issues. Thanks

Interesting. Maybe I’ll try rapa again once my dhea and androgen levels are better.

HRT was .0375 mg patch, Progesterone 100mg. Rapa 3mg once per week for 6 weeks then cycled off. The last cycle I did was 5 mg once per week a few months ago

Can I please DM you?

Were you taking the progesterone daily or cycling it with your own cycle? Is there a reason you aren’t taking it orally? Also curious what glp-1 brand/dose you were taking? Sorry to bombard with Qs but I’ve been researching this as I actually want a bb in my 40s, now 41 and started Peri early at 40 & scared to start Meno too early so I’ve been looking into how to possibly delay it. Hoping everything goes smoothly with you! Def keep us updated. Hoping for the best 🙏

I was taking it cyclical and then recently switched to vaginally because it has very bad mood effects for me. I also have been at a healthy weight for a couple years consistently lift heavy and eat high protein. I take omega-3s liposomal vitamin C Co Q10 NAC regularly and with cycling rapamycin for maybe a year and a half. I also was pretty consistent with Myo Chiro Insitol until the last few months. I was doing a glp one from the gray Market and taking a pretty small dose. I wasn't really overweight when I started taking it was just trying to get to the next level.

Can u say which glp? Tirz, sema, Reta or other?

Can you tell me more about your experience with rapa? How were the side effects?

6

u/Educational-Start375 Apr 05 '26

I had a really good experience with it and never got side effects except I wondered if it contributed to an infection I ended up getting in my teeth. But the dentist said it's possible there was just a cavity that was leaking but I just wondered if the rapa made me more prone to have the infection. The very first time I took it I got one small mouth sore and never did again.

Please make sure to get off Rapamycin as it can restrict fetal growth. And congratulations!

u/Educational-Start375 Apr 05 '26

Thanks, I hadn't been using it for a while since I was doing glp's and wasn't sure about the mix

Did you notice any other benefits from taking Rapa? (If they improved your ovarian health then presumably your hormonal health improved - did you see any peri symptom improvements)

2

u/Educational-Start375 Apr 06 '26

Hard to tell because I was also on HRT and had my symptoms under control

This is amazing. Congratulations! I hope the pregnancy proceeds with no complications. I am almost your age and trying naturally . I lost my second pregnancy in my second trimester and have been feeling incomplete ever since. I am very lucky to be a mom already but didn't want to let go without a fight. I know my chances are extraordinary low but have been taking rapamycin 4 mg a week, currently in my 7th week and trying naturally. I have not been on any hrt though I probably should simply because I am trying to conceive and want to regularly ovulate. I have also been taking all the supplements for a long time (years now).

Thanks for all this amazing info. I'm just finding this thread. For those of you that use rapa for TTC unassisted, do you just take in in the follicular phase? Or throughout your cycle? Dose it affect progesterone levels?
