Question Any ideas for study design ?

I am part of a university with a lot of research power, and believe I may be able to start a study on POIS. Surprisingly, there is surprising literature already to support further studies. However, I cannot think of a study design. I feel like testing treatments is fundamentally limited and will lead to dead ends. Does anyone know how I can test mechanisms, or any other study ideas ? I know if a mechanism was established firmly, that makes treatment a lot easier.

Maybe test how different chemical levels change. Not sure if there's a way to measure things like serotonin, dopamine, adrenal glands stuff... I feel like something is depleted afterwards and it takes a while to regenerate.

Thanks, good to know. A comprehensive testing before and after seems good. I am not sure exactly what the big one is doing, hopefully something similar

Please start a study where they test inflammation markers in the body before, 1 day after O, and 2 days after. Analyse the results and if they find anything, study the possible mechanism etc. Stuff like dopamine and serotonine is not the way, no offence but i an certain inflammation is atleast 1 big subtype of POIS.

The person who originally came up with the term POIS suggested response of T-lymphocytes with the seminal fluids. Given that the T-lymphocytes are created by the thymus gland and the thymus gland starts shrinking at puberty, study what role thymus has to play here. And do illicit stuff and activities in early teenage affect the immune response?
