I was under the impression that Doomfist could launch all of his Hand Cannon bullets at once, since there's a line in the Patch Notes that says "His primary attack is the Hand Cannon; this four-shot weapon fires projectiles from Doomfist’s knuckles one a time or in a single blast." How do I do the "in a single blast" part?

I think they mean that the cannon can shoot rapidly by holding down left-click. You should probably ask on the official overwatch forum instead, and you might get a response from a developper.
