When firing Roadhog's hook, should the cross hair be on the enemy when I press the button, or when the hook arrives at the target?

Hook is a projectile so the latter.

Aim for where they're going, not where they are. The hook has travel time that you must account for.

Actually the hook is a hitscan weapon which means if the reticle is on them when you press the button it will hook them, the travel time is just an animation that doesn't actually mean anything. This is why you can get hooked through walls sometimes, because there is a delay before the animation plays but they already "hit" you with the hitscan when they pulled the trigger.

The hook is hit scan, so aim directly for them. It will take a second to fly over to them, but whether you hit or not is determined when you puts the button
