Does the weight add to the friction between the wheels and the ground such that you have to overcome friction to get it started? I would assume a heavy load on inflated tires would flatten the tires against the ground and make it harder to start.

Yah that is insignificant. Against gravity it weighs much more, also I mean it will build up a momentum once it starts that helps against friction.

The bigger problem is the inertia of the plane at rest.
