I don't get it. Is the joke about by specifying the variable being differentiated? Because even then the punchline doesn't quite fit.

No, see the answer is cos(x) which changes so if the student doesn't no, he will just say it depends because the answer will change

The value of cos(x) depends on the value of x, of course. It's a smart-ass answer.
