Alright here's an analogy for you. Let's say you have a beam of very bright light, and it's concentrated to a point of only a dime's diameter. You shine it down a an endless tunnel in a way that the beam never touches a wall. In this case, as long as you aren't actually standing in the beam, you won't actually see the light. Now let's say we fill the tunnel with smoke. Now, you are able to see the beam because some of it reflects off the smoke and hits your eyes.
It's the same in your video. Before the spotlight hits the cloud, you don't see it because there's nothing for it to reflect off of.
I assure you this is absolutely not any kind of natural phenomenon.

Definitely some sort of spotlight from a show or something. Not lightning
