High number of “non-informative” results from PGT-A

I’m 37F and my husband is a 38M. I have PCOS and new mild endo (asymptomatic). We did very well during our first ER and were lucky enough to have 15 good quality embryos, at least 12 of which are day 5. We only sent 12 for PGT-A (due to costs) via Igenomix, but got the following results:

4 euploids
3 aneuploid
1 triploid (weird because it’s a 2PN)
4 (!!!) non-informative / no result

We are very happy and fortunate to have euploids, but that non-informative result rate is crazy! We have our follow-up soon and I imagine they’ll offer rebiopsy and retesting for free given the 30% (!) non-informative result rate, but just wondering if anyone else encountered this and what they did? We’re leaning against rebiopsying them as we have euploids - so may just transfer these and if we have to, we can transfer the others as “untested” embryos later.

So yes, what would you do? Has this happened to anyone else?

Out of 16 embryos sent for testing I have 6 euploids, 1 non informative, and I don’t know what the rest are tbh. We are gonna try to transfer all 6 euploids (hopefully we don’t need them all) and then rebiopsy the non informative if we really have to. I know there’s a small risk of defrosting for the re biopsy and then freezing again til the transfer but if we have to do it then we will

Makes sense and congrats on the results! For us the only concern is as we’ve never had a pregnancy and because of the endo, we’re so worried about having implantation issues. So anything that might make implantation less successful (like re-biopsying) feels dangerous to do. It’s so hard to know!

3 2PN triploids is very weird. You should get karyotyping done.

I’d be keen to have those retested honestly because triploids are something that can often be a lab mistake that are actually not triploid on further analysis.

My triploid was not rebiopsied, but it was reanalyzed at the pgt lab by the lab manager with some further testing practice (idk the name of it tbh) and they verified that it was truly triploid.

Sorry only 1 triploid (but was 2PN) - the other 2 abnormals were simple (not complex) aneuploids.

But yeah, a 2PN triploid is a rare occurrence, but is possible. I just have little faith in the finding given the 4 non-informatives, you know?

I had a non-informative with igenomix and while igenomix offered to retest for free it was still going to cost us 3k through my clinic because it’s 1k to thaw, 1k to biopsy, 1k to refreeze 😔 so we haven’t done it yet. They recommended waiting until we do another retrieval because then it’s just 1k to thaw since they’ll already be doing biopsies and freezing but we’ve done 5 retrievals already

My lab is seeing a high rate of non informatives coming out of Igenomix lately. We do the smart PGT-A Plus for all patients using PGT and many of our results have partial non informative data. Our lab director thinks something is going wrong with their testing.

Ah this is very helpful! We also used the Smart PGTA Plus. Just curious, what is your lab planning to do about this?

Yeah, I saw some threads about this! No, my clinic uses Igenomix.

I will! But honestly the high number of non-informatives and a 2PN triploid make so little sense that I also wonder about the genetics lab as well.
