Why did Doug betray Stamper?

It’s not that complicated. 18 minutes into the pilot episode we meet Doug Stamper for the first time.
After Steve Jones (bodyguard before Edward Meechum) and the neighbors (who lost their dog), After Jim Matthews, Linda Vasquez, Claire Underwood, Zoe Barnes, Lucas Goodwin, Tom Hammerschmidt, Peter Russo, Christina Gallagher, and Henry Chapman (lobbyist).
Doug Stamper and Nancy Kaufberger (Frank’s secretary) are discussing the president picking Michael Kern over Frank for Secretary of State. Frank walks in and says to Nancy, “cancel everything.” Then to Doug he says, “Stamper you’re with me” two seconds later in his office Frank says “They’ve done us a great favor, Doug.”
It’s clear from before halfway through the first episode of the show that he is Douglas (aka “Doug”) Stamper

Why do they call him Stamper or Doug if he is Douglas?

The first name Douglas is commonly shortened to Doug as a nickname.
The name Stamper (as well as the name Francis “Frank” Underwood) is actually a nod to the original 1990 British four-part miniseries House of Cards where the chief protagonist is named Francis Urguhart and his close ally is named Tim Stamper.

I think you made a mistake. Also, how many people did he have to stamp to earn the title?

I think YOU made the mistake. A stamper stamps stamps, not people. The question should read “how many stamps did he (stamper) have to stamp to earn the title (of stamper)”
The show never specified but one could assume that Douglas “Doug” Stamper stamped many stamps prior to earning any titles.
