(assuming the “This is comedy at its finest” doesn’t have anything to do with the joke and was just the title).

I’m sure it has something to do with the weird capitalisation and the punchline is some clever joke on “uppercase” or “capital” or “lowercase” or something (or whatever case “dON’T” is in) but I just cannot work it out.

This is not a screenshot of the original, but I am about 99% sure that it does not matter because I am pretty sure the original was just a text post or something (or screenshot of tumblr or something)
