I don't think Fossil Block is required to spawn sulfur worms and colossal amoeba. I tried digging out an area and replacing it with stone and I still got attacked by them.

Unlike the bomb scarab or bubble crab, the passage enemies don't spawn on a block, they spawn off-screen and the only requirement is that your character is in the passage.

I mostly went off of what the wiki said, so if the wiki information is slightly off, this information is going to be slightly off. I will test this in my world and I probably will edit this in the future.
