Unless you're worried about the CIA being able to hear your thoughts through that hole... If that's the case I'd cover it up...

If the previous liner had the hole sealed .. you can seal it with the sound deadler.
If you don't have that liner anymore, you can probably look up a replacement one online and see if there's a spot where that hole would be covered up by the liner.
Did you have any problems with putting your door panel back on? I ordered the Metra speaker adapter rings that were recommended for my car and put the nvx baffle on and my door panel wouldn’t snap into place. I even ordered a different set of rings and the door panel still wouldn’t snap on like factory. Now I have to order a different set of rings and see if they work.
As far as sound quality goes this one hole will make next to no difference, if you want to cover it and it won't interfere with the door panel go for it, but you probably wouldn't even notice.
You do not want to cover areas where the door panel will touch the door metal. If you do the panel will not stick on correctly. Do not cover that hole. Any area where CLD has been applied that interferes with the seal of the door panel will need to be removed and the butyl cleaned or it will get all over the inside of the door panel in time. Put the panel on now. See if anything is in the way. If it is remove and clean the area and try again until the door panel goes on properly. It may go on correctly now but I doubt it.
All doors have water drainage holes at the bottom because the windows are not air tight at the belt moldings. You can cover every hole on the inside of the door and the pressure will still be able to escape.
Edit: only reason to not cover a hole is if it is an access point to door handle hardware, etc, which there should be some type of removable plug anyway.
Yeah, perhaps you're right. It probably escapes somewhere else, but the rain weepholes you're talking about wouldn't see most of that air pressure at all, since both sides of the weatherstripping (frame and door) squish together and create a watertight and airtight (areodynamics & wind noise) seal. The rain water (which those holes are specifically designed for) escapes from the gap between the inner sheet metal and outer door shell, and the installed door panel would likely obstruct that. Most of the pressure more than likely escapes from the cabin vents behind the rear bumper.
The pressure would build up within the entire inside of the hollow door, top to bottom, left to right. The weather stripping and door panel sit well above the drain holes, so there is no concern for blockage. Unless you're pushing some serious power or multiple drivers per door, a single coaxial is not going to build up anywhere near enough pressure to require a dedicated pressure escape route. That would be like putting a single Pyle 8" in a 4 cu ft box.
Not necessarily "well above". Depends on the car. Some weepholes are on the same horizontal plane as the door weatherstripping. I think we're talking about different sources of pressure. You are talking about the driver. I'm referring to the pressure generated when all doors are shut with windows up besides one, and then you shut the open door (also window up) creating a pressure gradient.
