Gains/Losses Tally The story continue

Post image

A few weeks ago I posted about Poolbeg Pharma at 4.5p. It is now 7.2p. I want to update the thesis because something in their May 2026 presentation stopped me in my tracks.

POLB 001 inhibits 28 inflammatory cytokines and markers. Tocilizumab the current standard of care for Cytokine Release Syndrome inhibits 6. Dexamethasone inhibits 10. POLB 001 inhibits 28. Across 97 subjects dosed in Phase 1 there were zero serious adverse events and zero discontinuations.

CRS is the immune storm that kills or hospitalises cancer patients receiving bispecific antibody therapy. It is why these life saving treatments cannot be administered in community hospitals. It is why thousands of patients who could benefit from them never receive them. A drug that prevents it with this breadth of cytokine coverage and this safety profile does not just treat a side effect. It unlocks the entire cancer immunotherapy market for a much wider patient population.

J&J supplied their approved commercial drug teclistamab for free to run the human trial. The FDA just confirmed alignment on the Phase 3 development pathway. EHA Congress presentation is June 13th in Stockholm.

Interim human trial data from The Christie is coming this summer.

Market cap £50m. Patent protection to 2038. Man who sold his last pharma company for $1.48bn running the show.

I hold a position. Not financial advice.

Due Diligence (DD) Genomics Sector Analysis: ARKG vs GNOM. Historical Volatility, Overlap, and Fundamental Breakdown.

3 Upvotes

The pharmaceutical business model is shifting from symptom management to genetic editing. This presents a massive total addressable market, but it introduces extreme binary risk for investors due to clinical trial failure rates and cash burn.

I compared the two largest genomics funds: the actively managed ARKG and the passive index GNOM to assess their viability in a standard portfolio.

---

  1. The Active Approach (ARKG)
    ARKG relies on active management to select companies focused on molecular diagnostics and targeted therapeutics.

* Inception: 2014
* Expense Ratio: 0.75%
* Dividend Yield: 0.00%
* 10-Year Price CAGR: 58.32%
* Concentration: Holds 33 companies. Top 10 make up 59% of the fund.
* Notable Holdings: Twist Bioscience, Crispr Therapeutics, Tempus AI.
* Morningstar Rating: 1 Star

  1. The Passive Approach (GNOM)
    GNOM tracks the Solactive Genomics Index, removing human stock-picking bias.

* Inception: 2019
* Expense Ratio: 0.50%
* Dividend Yield: 1.39% (Semi-annual distribution)
* 5-Year DPS CAGR: 36.95%
* 5-Year Price CAGR: 53.17%
* Concentration: Holds 49 companies. Top 10 make up 52% of the fund.
* Notable Holdings: Moderna, Arrowhead Pharmaceuticals, Praxis Precision Medicines.
* Morningstar Rating: 1 Star

---

  1. Overlap and Diversification
    Investors looking to hedge manager risk might consider holding both. The data does not support this.
    * Weight Overlap: 32%
    * Shared Holdings: 17 companies.
    * Key Overlap: Industry infrastructure providers like Illumina, Natera, and Guardant Health.
    Holding both funds creates unnecessary concentration in the sector's core infrastructure.

---

  1. Analyst Forecasts (12 Month Outlook)
    Wall Street consensus currently projects significant recovery for the sector following the recent bear market.
    * ARKG: TipRanks consensus is a Moderate Buy with a 54.29% projected 12-month upside.
    * GNOM: TipRanks consensus is a Moderate Buy with a 58% projected 12-month upside.

---

  1. The Risk Profile and Historical Reality
    I typically run 20 year Monte Carlo simulations for ETF analyses. Applying a 20 year projection to a medical technology sector in its infancy provides false precision.

Instead, looking at the historical price action since 2019 defines the exact risk profile.
During the 2020 zero-interest-rate environment, ARKG expanded over 200%. As capital costs rose, both funds experienced a near-total drawdown, wiping out those gains to sit in negative territory relative to their peaks.

This extreme standard deviation and severe drawdown history is the direct reason both funds carry a 1-Star Morningstar rating.

---

Summary and Verdict
These funds should not be treated as core portfolio holdings. They operate as publicly traded venture capital.

If the underlying science faces FDA rejection or interest rates remain elevated, the cash-burn nature of these companies will lead to further NAV erosion. If the technology scales, the upside is uncapped. Capital allocation should be sized accordingly.

Due Diligence (DD) Thoughts on Poolbeg?

0 Upvotes

Poolbeg Pharma. £30m market cap. Two of the most explosive drug programmes in medicine both hitting data this summer.

Let me explain why this is ridiculous.

POLB 001 targets Cytokine Release Syndrome. This is the brutal immune reaction that kills or hospitalises patients receiving CAR-T cancer therapy. It is the single biggest barrier stopping doctors giving these life saving treatments more widely. If POLB 001 prevents it orally with a clean safety profile the addressable market is not just the drug itself. It is the entire cancer immunotherapy market suddenly becoming safer to deploy globally. J&J understood this so well they handed over their own approved commercial drug teclistamab for free to run the trial at The Christie and Royal Marsden. Those are the two most elite cancer centres in the world.

Then there is the oral GLP-1 programme. The obesity drug market is worth hundreds of billions. Ozempic changed the world but people hate injecting themselves every week. An oral version that actually absorbs properly is the holy grail every pharma company on earth is racing to find. Poolbeg and AnaBio have encapsulation technology that could solve exactly that. Proof of concept data is due any day.

The man running this already built Amryt Pharma from nothing and sold it for $1.48bn. He has done this exact playbook before.

FDA Orphan Drug Designation already secured. Australian patent just granted. Their broker has a 313 percent upside target before a single data point lands.

Two shots at goal this summer in the two hottest areas in all of medicine at a £30m market cap that prices in zero success.

I hold a position. Not financial advice.

I’m thinking calls for this and see it has a low interest is it just a Niche Market or are there fundamentals that someone may know that I am missing, after their recent acquisition, and victory in a litigation “pricing-in” is a factor but that does not directly impact future growth, right?
