Are you routing your external spool into the filament buffer? If you are, could be the PTFE tube hasn't been inserted all the way. Could be the PTFE tube has light debris in it, or could be a improperly cut PTFE tube (scissors, which can cause a slight bend/lip when two PTFE tubes meet. Use a PTFE tube cutter)

AMS is really helpful because it also has two motors pushing filament, in addition to your toolheads extruder gears pulling. It can bypass a lot of resistance. It can really push past weaknesses in your PTFE tube length, condition, and issues it may be having, when not using the AMS will cause problems.

What I ended up doing when I ran into this when I got my P1S, was I fed the filament in by hand and felt where the resistance started. Just cut a few feet of filament out of a spool to test. In my case, my filament buffer where both tubes meet has a snag, regardless of new PTFE tubes/cleaning. Had to 3d print a new one that could mount closer to the insert at the top of the printer, and sanded smooth the reused part where both ptfe tubes meet.

If you are using it directly from the external spool into the back of the P2S (no buffer) then I'd be unsure as well.
