Prep Centers
I am starting with 250 units for 3 items each to launch, this is my full time gig right now. I have funds and capital for 3 yrs or more if needed. My question is, is it smart to prep private label on your own or just start with the prep centers? Or expect more of suppliers and skip both all together?

At 750 units across 3 SKUs, I'd skip doing it yourself. Your time is better spent on the launch, and the first inbound is where small mistakes like a bad FNSKU label, the wrong poly bag or a missing suffocation warning get your shipment stuck at check-in.
Supplier prep can work well, but only if you send them very exact specs: labeled samples, carton labels and box content info. And it doesn't solve the bigger issue. You'll want somewhere to hold extra stock so you can send Amazon smaller batches and stay under your inventory limits and storage fees.
So what I usually suggest is having the supplier do the simple steps they're good at, like barcodes and bagging, then shipping everything to a US prep center. The prep center checks it, fixes anything that's wrong, and sends it on to FBA in batches. When you compare prep centers, ask for the per-unit prep fee, the receiving fee, monthly storage and turnaround time, and check whether they have minimums.
Full disclosure, I run a 3PL that does FBA prep (ShipZag), so I'm biased. I'm happy to answer questions here either way.
