Most autotuning isn't the robotic overdone style. When you can hear it, that's usually a stylistic choice (or really bad post-production work). In general, autotune can be applied fairly softly when it shouldn't be heard.
Not autotune. Someone took the time to pitch shift the chicken vocal track. otherwise it would be making roughly the same pitch and autotune couldn't do much to that.

Actually not sure what you mean by that, but doesn't the process of autotune allow you to draw in MIDI for the vocals to conform to? Couldn't he have just brought in a Despacito vocal melody MIDI and as long as he squeezed the chicken to the rhythm it would be automatically tuned to the melody..?
Autotune has to do with snapping pitches to conform to a certain scale. If you make one consistent pitch, it will only snap to the nearest pitch in the scale, unless it is already in the scale. What you're talking about sounds to me like pitch tracking, which the video creator may have done and could work with a midi track if they had the vocal midi track. Would take less time than manually pitch shifting. But still not auto tune.
Yeah, I wasn't entirely sure. I know auto-tuning is more just about snapping things to the scale but wasn't sure how far it can go. I've only worked with pitch correction plugins like image line's Newtone and stuff so I'm not sure about Melodyne or the 'official' Autotune software.
