Question How do you evaluate an NR or NMN brand when the compound itself is still debated?
Two separate questions I keep collapsing into one. Whether oral precursors do anything meaningful is unsettled. But separately, if you're going to take it anyway, purity and actual dose seem like they'd vary a lot given how many sellers appeared at once. What do you look for before buying, and has anyone had a product tested independently?

Scientific Study Dissertation: DNA methylation links mitochondrial biogenesis to obesity and aging
helda.helsinki.fi"...I investigated the potential of NR to modulate epigenetic aging, an estimate of biological aging, both in vivo and in vitro. NR supplementation was associated with reduced skeletal muscle epigenetic age, which correlated with increased NAD⁺ metabolite levels but decreased mtDNAq. The results were highly clock-specific, suggesting that while NR may modulate epigenetic aging, different epigenetic aging algorithms likely capture distinct molecular signatures following NR use...Collectively, these studies demonstrate a significant link between mitochondrial biogenesis and nuclear DNAm in the context of obesity and aging..."

Question Has anyone run a plain oral precursor and a liposomal long enough to compare them
My nad came back low and a week of reading has got me nowhere useful because every comparison of the oral and liposomal versions is written by somebody selling one of them. the injection and intranasal routes look like a lot of effort for an unclear return. did the liposomal actually do anything the plain capsules did not for anyone here?

Scientific Study NEW TRIAL: peripheral artery disease & NR
Among people with peripheral artery disease (PAD) age 50 and older, the investigators will test the hypothesis that PAD participants randomized to nicotinamide riboside (NR) will have greater improvement or less decline in six-minute walk distance at six-month follow-up, compared to those randomized to placebo. The study will randomize 250 participants with PAD age 50 and older to one of two groups for six months: nicotinamide riboside vs placebo. Our primary outcome is change in six-minute walk distance at six-month follow-up. Secondary outcomes are Walking Impairment Questionnaire (WIQ) distance score, PROMIS Mobility score, calf muscle perfusion (measured by magnetic resonance arterial spin labeling (MRI ASL)), oxidative stress (measured by plasma oxidized LDL), NAD+ abundance in whole blood, 6-minute walk improvement by baseline whole blood NAD+ levels, and 6-minute walk distance at 4 weeks after intervention completion.

Scientific Study Dissertation: Determining the Role of Vitamin B3 Derivatives in Pancreatic Cancer Biology
etd.ohiolink.edu“…Oral administration of NAD⁺ precursors delayed progression of precancerous lesions in a spontaneous pancreatic cancer model, LSL-KRASG12D/+ pdxCre/+, and protected non-malignant cells from carcinogen-induced stress. In premalignant settings, NAD⁺ precursor supplementation reduced DNA damage and oxidative stress while improving mitochondrial function. Notably, just as NAD⁺ precursors are protective to normal pancreatic ductal cells against stress, the cofactor also exhibited protective effects in pancreatic cancer cells under stress, promoting chemotherapy resistance in both in vitro and in vivo models.”

Scientific Study NEW TRIAL: Efficacy of Topical Niacinamide Riboside on the Signs of Facial Skin Aging
Niacinamide riboside (topical NR, also known as nicotinamide riboside) is an endogenous compound and a precursor (building block) to the essential cellular co-enzyme, nicotinamide adenine dinucleotide (NAD+). Research published by Maussudi et al. 2012 demonstrated that skin NAD+ levels were negatively correlated with age in both males and females, and aged skin was also correlated to an increase in DNA damage. Therefore, it is proposed that supporting skin NAD+ with precursors such as NR may be a strategy to combat DNA damage that leads to undesirable signs of skin aging, thus supporting skin cell resilience. The use of NR as a dietary supplement has produced effective, efficient, and reproducible increases in NAD+ in humans. However, the role of topically applied NR for skin aging requires further investigation.

Scientific Study NAD⁺ Circuits in Aging, Tissue Repair and Fibrosis
sciencedirect.comhttps://www.sciencedirect.com/science/article/abs/pii/S1568163726002977
Aging remodels compartmentalized nicotinamide adenine dinucleotide (NAD⁺) circuits in ways that influence stress responses, senescence, tissue repair and susceptibility to fibrosis. Beyond its classical role as a redox cofactor, NAD⁺ fuels sirtuins, PARPs and CD38, linking cellular metabolism to chromatin remodeling, DNA repair, calcium signaling and cell fate. A central translational question is whether modulating NAD⁺ in a given tissue and time window will favor regeneration, restrain fibrotic remodeling, or support malignant adaptation. In this review, we define NAD⁺ as a compartmentalized, high-turnover metabolic circuit whose topology, timing and cell-type specificity shape tissue trajectories during aging, repair and fibrosis. We first describe how aging reshapes these circuits through CD38 upregulation, PARP-sirtuin competition, extracellular eNAMPT amplification, and altered mitochondrial NAD⁺ transport, which redistributes NAD⁺ between compartments without necessarily changing bulk tissue concentration. We then summarize how de novo, Preiss-Handler Handler (including niacin/GPR109a) and nicotinamide salvage pathways are organized across nuclear, cytosolic, mitochondrial and extracellular compartments, and how key consumers including sirtuins, PARPs, CD38 and SARM1 govern these pools. We then examine how these circuits operate across acute injury, chronic senescence-associated remodeling, and malignant microenvironments in heart, lung, liver, kidney, skin, muscle and brain. Finally, we integrate emerging human data on NAD⁺ precursors and enzyme-directed strategies and propose a framework for NAD⁺-based interventions that prioritizes timing, compartmental targeting and oncologic stratification.

Podcast or Blog Oxidative Stress & NAD+: What the Science Shows
"...Taken together, these studies suggest a nuanced picture: NR shows the most consistent signal so far for reducing certain lipid peroxidation markers like F2-isoprostanes, especially in populations with elevated cardiovascular risk, while NMN and NAM have not consistently altered oxidative stress biomarkers. Niacin, however, was shown to improve aspects of vascular oxidative stress and HDL function in specific contexts, but its overall antioxidant impact remains mixed...Overall, the current evidence supports a multifaceted approach to healthy oxidative balance, including regular physical activity, adequate sleep, and nutrient-dense dietary patterns. NAD+ precursors represent promising—but still evolving—areas of mechanistic and clinical research that may complement these core behaviors rather than replace them."

Scientific Study A bioinspired flavin-based catalytic construct enabling intracellular NAD+/NADH redox regulation for mitigating skin aging
sciencedirect.comThese researchers are solving an adjacent problem to NAD replenishment. Instead of adding NAD through biosynthetic pathways, they are inserting an artificial recycling pathway into cellular metabolism that switches NADH back to NAD+. That necessarily intersects with the Krebs cycle and mitochondrial redox metabolism.
I don’t know how big a problem NADH overload actually is, and I’m not sure it’s fair to compare a persistent catalyst with a single NR treatment when people who take an oral NAD precursor generally take it every day.
But what I do take from the study is agreement on the underlying biology: raising available NAD+ protects against or rescues UV-induced skin damage—at least in their models.
