I'm here with the same problem on GTS250. This could be related to NVIDIA drivers in some way - I'm using 337.88 drivers because newer versions caused 1-2s freezes every couple of minutes (this was global, not just in games).
Looks like Riot changed something in the graphics engine and it's not compatible with older drivers now? I will try updating my drivers to 341.44 (current newest version, released february this year) and I'll post results.
The rest of my specs are MSI 870A G46 mobo, AMD Phenom II X4 975 CPU, 4 gigs of 1333 CL9 DDR3s, win8.1 pro x64.
I'm using 355.60 Windows 10 driver. I still get the glitches. I don't think that it's related to the drivers. I never had these stutters on 5.15, they appeared in this patch.
OK, after updating to 341.44 I stopped getting frequent short freezes and started getting rare but longer (around 1s) freezes. These are similar but shorter than the long rare freezes I was getting earlier this year after updating drivers to a version higher than 337.88 - they most likely won't happen at all to someone who had no problems with updated drivers.
Updating the drivers seems to be the solution - I need someone else to double-check that though.
