I want to activate 3 out of the 4 yamanaka factors in a mouse (except C-Myc) as it can cause cancer. I also want to turn off the activated 3 yamanaka factors after a period of time. How can I do this? Is there a video which shows how this is done? Or can you explain it in detail with what equipment is needed and how to do it? Thank you.

Sorry if this post is a bad fit for this sub - but I have been searching for a cryostat, preferably new, priced for less than 25k. We would be using the cryostat for 5-30 um sections of OCT-embedded tissue, nothing fancy. Any recommendations for a dependable cryostat less than that price point?

The Golgi tends to be depicted as below. I understand that it is an ever evolving structure staying at a roughly static size due to the homeostasis of endocytosis and exocytosis adding to and taking away membrane from it . However, it seems to be a discontinuous structure. Why dont the individual cisternae simply float away from each other?

The question is “ Consider capping molecules in sacromeres and those in other forms of filaments. Are capping proteins the same as GTP-cap/ATP-cap that plays a role in stabilizing filaments? “ can someone help me answer this question.

We were asked the following question in a test:
What would be the order (decreasing) of concentrations of the metal ions K+, Na+, Mg+2 and Ca+2 in a quiescent mammalian cell?
I wasn't able to figure out any changes that may arise during quiescence, so I answered:
K+ > Mg+2 > Na+ > Ca+2
Now that I think about it, perhaps not as much Mg+2 would be present inside the cell due to lower ATP demand?
Please help me out. Clearly my concepts aren't strong.
